In this study,the complete mitochondrial genome sequences of 6 genera and 6 species of formicidae(Cerapachys xizangensis?Myopopone castanea?Tertaponera allaborans ?Myopias luobaensis?Doxrylus orientalis?Amblyopone crenatum)were determined by high-throughput sequencing,and some high-throughput sequencing gaps were supplemented by self-designed primers.For each kind of the mitochondrial genome of annotation and analysis of the bases,and download has been published on NCBI formicidae 20 species of mitochondrial genome sequences,extract protein genes and rRNA genes and the joint of 6 species,this study established the COX1,COX2,rrnL,rrnS,COX1+ COX2 protein gene 6 + rrnL + rrnS and data sets,build the BI trees and ML on analyzing phylogenetic tree,aimed at exploring formicidae the species in the division level and order the phylogenetic relationship on the yuan,enrich its mitochondrial gene database.The following results were obtained in this study:1.The mitochondrial sequences of crenatum(Cerapachys xizangensis?Myopopone castanea?Tertaponera allaborans ?Myopias luobaensis?Doxrylus orientalis?Amblyopone crenatum)were 16769 bp,15890bp,15459 bp,15227bp,17792 bp,and 16016 bp.Among them,mitochondrial sequences of Myopias luobaensis and Doxrylus orientalis may have gaps near the CR region,and the rest sequences have been tested.2.It was found that mitochondrial protein genes of the 6 species were in the same sequence,and there was frequent gene overlap between atp8-atp6 and nad4-nad4 l.tRNA genes have more rearrangement of the phenomenon,such as CR area to NAD2 genes,between three genes(tRNA-Met,tRNA-Gln,tRNA-Ile)have active gene rearrangement events,tRNA-Ala gene in the mitochondrial genome of Cerapachys xizangensis and Myopopone castanea was inverted and tRNA-Val gene in the mitochondrial genome of Myopopone castanea?Tertaponera allaborans ?Myopias luobaensis?Doxrylus orientalis?Amblyopone crenatum was inverted,tRNA-Trp gene in the mitochondrial genome of Myopias luobaensis occurred ectopic inversion.It was found that there was a highly repetitive TATAAA sequence in the Intergenic region of Doxrylus orientalis.At the sametime,the NAD2 gene to the tRNA-Gln gene repeats.The NAD1 gene of Myopias luobaensis starts with TTG.3.In addition to the NAD1 gene of Myopias luobaensis,the rest of the protein genes all take the typical ATN as the starting codon,most of the protein genes take TAA as the termination codon,some of the protein genes take TAG as the termination codon,and a few of the protein genes end in incomplete code TA or T.By statistical analysis of RSCU of each species,it was found that the six codon types of mitochondrial protein-coding genes were mainly UUU?UUA?AUU?AUA?UAU?AAU,which was consistent with the obvious AT bias of mitochondrial genome.4.Most of the secondary structures of tRNA of 6 species were downloaded from the tRNAscan-SE website,and it was found that some tRNAs were mismatched,mainly U-U mismatched,and all of them occurred on the amino acid receptor arm.5.Use of COX1,COX2,rrnL,rrnS,COX1+COX2+rrnL+rrnS and PCGs data set set up six kinds of ML and BI trees,the results of polygenic joint contribution support the Formicinae and Myrmicinae as a sister group relationship,most accomplished results show that Doxrylus orientalis and Cerapachys xizangensis are very closely related,the results support the Cerapachyinae classified the Dorylinae.
