The mitochondrial DNA has been comprehensively applied for studying in population genetics, phylogenetic reconstruction, comparative and evolutionary genomics and molecular evolution, because of its simple structure, accelerated molecular evolutionary rates, maternal inheritance and lack of restructuring.In this study, the complete mitochondrial genome of Luehdorfia taibai Chou,1994, Paracymoriza distinctalis (Leech,1889) and Lista haraldusalis (Walker,1859) were sequenced, assembled, annotated and analyzed using the Sub-PCR technology. The six phylogenetic trees were reconstructed among33lepidoptera insects, using the maximum parsimony (MP), the maximumlikelihood (ML) and the bayesian inference (BI) methods based on the two rRNA gene sequence data and the thirteen protein coding gene sequence data. Conclusion were showed below:1. The mitogenome size of Luehdorfia taibai, Paracymoriza distinctalis and Lista haraldusalis are15553bp,15355bp and15213bp respectively. They all encode13protein genes,22tRNA genes,2rRNA and the A+T-rich region.2. The nucleotide compositions of mitochondrial genomes of Luehdorfia taibai, Paracymoriza distinctalis and Lista haraldusalis are biased toward adenine and thymine, and the A+T omposition of mitochondrial genomes (J strand) are81.47%,82.14%and81.51%, respectively.3. Most protein-coding genes have typical ATN initiation codon, except the COI gene of Luehdorfia taibai and Paracymoriza distinctalis is tentatively designated by the CGA codon as observed in other lepidopterans, and Lista haraldusalis use TTAG as start codon. Most Protein genes use the typical and complete stop codon:TAA or TAG, except the COII gene of Paracymoriza distinctalis and the COII and ND6gene of Lista haraldusalis, which use the incomplete stop codon:T.4. All the22tRNA genes of mitochondrial genomes have a typical clover leaf structure except tRNASer (AGN) of Luehdorfia taibai and Paracymoriza distinctalis, which lack the DHU arm. Some unmatched base Pairs occur in the tRNAs of Luehdorfia taibai, Paracymoriza distinctalis and Lista haraldusalis. Otherwise, the Luehdorfia taibai mitogenome contained an extra tRNALeu, located from191bp to259bp, of which function is unclear.5. The A+T-rich region of Luehdorfia taibai, Paracymoriza distinctalis and Lista haraldusalis are939bp,344bp and310bp respectively in length with the AT content up to94.56%,95.06%and96.13%. And the region included some typical structures of other lepidopteran mitogenomes, for example, microsatellite-like repeating sequences (AT)n and (ATT)n elements, poly-A and poly-T structures etc.6. The phylogenetic trees were reconstructed. The conclusions were drawn as follows: the grouping of (Noctuoidea+(Geometroidea+Bombycoidea)) formed a sister group always; Ctenoptilum vasava as the only representative date of Hesperoidea, have closely relationship with Papilionoidea; Saturniidae and Sphingidae have a closely relationship.
