Resting-state functional connectivity MRI is a novel non-invasive method to measure the BOLD signal.Due to the merits of convenience and flexibility and the great utilization potentiality in clinical, the technology is improving the traditional invasive medical testing, such as Wada test and cortical electrical stimulation. The language network in human brain is unique.Functional magnetic resonance(fMRI)technology was applied to study the language network at resting-state,revealed the connectivity and cerebrum asymmetry between brain regions and two hemispheres. In this paper, six steps were fufiled by using resting-state fMRI data with the help of several kinds of software under Linux and Window. The steps were pre-process,analysis of structure MRI(sMRI),analysis of functional MRI(fMRI),registration between sMRI and fMRI,analysis of connectivity and lateralization, and result display methods. In pre-process,SPM8 was employed in slice timing correction, rigid body correction for head motion, smooth and normalization.DPARSF was employed in low-pass temporal filtering, head motion regression, whole brain signal regression, and ventricular and white matter signal regression.Structure data were processed using FreeSurfer software package based on the Linux platform.Structure and functional images were aligned using boundary-based registration within the FsFast software package. Finally, using MATLAB software analyzed the processed data, and then doing the functional connectivity research. The final results displayed through freeview.Otherwise, using the quantitative intrinsic laterality index to measure language network asymmetry of human cerebrum at resting-state,and the affection of hand factor to asymmetry. The findings are:(1)The left-lateralized regions involved the inferior frontal gyrus, superior temporal gyrus, and temoral pole. The most right-lateralized regions involved the insula and angular gyrus;(2)Right-handed individuals showed stronger asymmetry than left-handed individuals. It is concluded that specific regions of the human cerebrum which associated with language network are functionally asymmetry at resting-state.Left-handed and right-handed participant show the asymmetry on the language network, and Right-handed individuals showed stronger asymmetry than left-handed individuals. It was implied that the hand factor have effect on the degree of language network asymmetry. This work might be helpful to show evidence to deal with medical information on clinic and provide basic research on functional integration of language network of human brain.
