The driving force of life is a physiological process in which various chemicals take part.Even a small presence of compounds can play a significant role.Metal ions are vital since they can maintain the normal structure and function of macromolecules such as protein nucleic acids and hormones.High concentrations of trace metals can produce specific toxic and pathogenic effects on human bodies.If those metals exist in the outside world,they may cause environmental pollution and may also enter the human body in a certain way,resulting in significant fluctuations in the body content.Thus,developing the in-cell metal content detection,detection of the initial phase of vital signs changes have an essential guiding significance of control and improvement of the status of the body.Compared with the traditional methods for determination of metal ions,the fluorescent molecular probe has the ability of high selectivity,high sensitivity,convenient operation and real-time in situ detection.Al3+and Zn2+,which are strictly related to human health,were selected as research subjects in this thesis.The corresponding probes were constructed and synthesized separately,their fluorescence properties were explored in the buffer solution,and the identification properties and detection capabilities were examined.Some characterization techniques were used to verify the mechanism.Chapter 1:The design principle,detection mechanism and developing status of small molecular fluorescent probes for metal ions are summarized.Chapter 2:Based on the principle of CHEF,the tetradentate ligand azo compound fluorescent probe HDBD with strong chelating ability of Al3+was constructed,which could be easily prepared by diazonium coupling reaction of one-pot synthesis using 2-aminophenol and resorcinol.The fluorescence intensity of HDBD(at?ex=467,?em=566 nm)increased more than 8-fold by binding to Al3+,and the probe is highly selective for Al3+.HDBD had been successfully used for Al3+imaging in Hela cells.Chapter 3:Based on the principle of ICT,the bipyridyl bidentate ligand fluorescent probe BPDO with fast coordination ability with Zn2+was constructed,which could be prepared by one-step synthesis with simple starting materials 3-hydroxypyridine and iodobenzenediacetic acid under room temperature.The fluorescence response of BPDO towards Zn2+showed good selectivity and the fluorescence quantum yield of BPDO-Zn2+complex was high.BPDO had been successfully used for Zn2+imaging in HepG2 cells.Chapter 4:Summary and prospect.
