As an important regulatory substance in plant growth,the role of plant hormones in the process of signal transduction and the regulation of stress response genes have been the research focus in recent years.In earlier studies,our research group cloned a inositol galactoside synthase gene(AnGolS2)from dongqing,xinjiang,and confirmed that AnGolS2 is an anti-stress gene that responds to a variety of abiotic stresses Therefore,in this study,bioinformatics analysis was conducted to compare the homologous Gol S proteins that had clearly responded to the low temperature response in other species with AnGolS2 proteins of solanopsis solanopsis in order to explore whether the differences in n-terminal and c-terminal of AnGolS2 proteins and cold-resistant Gol S proteins of other species were related to the cold-resistant function in the conserved domain.Meanwhile,QRT-PCR technology was also used to analyze the changes of transcriptional expression of AnGolS2 gene in different parts of plants under the induction of different hormones.Analysis AnGolS2 gene promoter non-coding regions hormone response signal of cis element and constructed three different promoter fragment missing plant expression vector,injection of tobacco leaf instantaneous expression and protein GUS staining in different hormone treatment after the experiment,clear response cis effect of different hormone signal components in AnGolS2 the role of expression of gene transcription,the experimental results of this study are as follows:1.By AnGolS2 protein bioinformatics analysis,we found that in several plants Gol S protein N-and C-not with the source area of inositol galactose glucoside contribute less synthetase gene function and in the middle of a AnGolS2 25-300 amino acid protein conservative district,the inositol galactose glucoside synthetase gene function,the contribution of this area is not only contains glycogen GT8 family function of protein structure domain and two conservative motif,or main secondary structure formation of the area,its structure formation and stability of Gol S protein and protein functions affect decision effect.2.In the root system of Ammopiptanthus nanus,ABA GA3 and IAA can significantly increase the transcriptional expression of AnGolS2 gene in the root system,while exogenous ET can significantly inhibit the transcriptional expression of AnGolS2 gene.In the stem of Ammopiptanthus nanus,exogenous IAA Me JA and SA can significantly increase thetranscriptional expression of AnGolS2 gene in the root system,and exogenous GA3 can negatively regulate the expression of AnGolS2 gene in the stem,while ABA and ET have no effect.In young leaves of Ammopiptanthus nanus,all the five hormones except SA can significantly positively regulate the expression of AnGolS2 gene,which indicates that different hormones have different effects on the transcriptional activity of AnGolS2 gene in different tissues of Ammopiptanthus nanus.these results suggest AnGolS2 gene transcription activity affected by hormones and tissue.3.Under normal conditions,the protein chemical staining showed that GUS AnGolS2 gene is a lower expression,however,under the different hormone induced,contains various cis acting element AnGolS2 promoter gene transcription activity of downstream report is different,and there are interactions between these hormones that suggests AnGolS2 gene promoter on the relationship between these hormones response element influence AnGolS2 gene transcriptional expression,promote AnGolS2 gene transcription under abiotic stress to improve plant resistance.
