The circadian clock is a metabolic,physiological and behavioral biological phenomenon in which the biological response to the cyclical changes of the external environment produces a certain rhythm.Recent studies have shown that in human cell lines,the circadian clock regulates intracellular magnesium ion concentration by regulating magnesium ion transporters,while intracellular magnesium ions regulate the circadian clock by regulating cellular metabolism and translational levels.In plants,Mg2+participates in a series of physiological and biochemical processes and plays an important role in the growth and development of plants.So far,little research has been done on the mutual regulation of plant magnesium nutrition and circadian clocks.In this paper,Arabidopsis wild-type col,Arabidopsis circadian rhythm core gene single mutant cca1,lhy,toc1 and over-expression material CCA1-OX were used as materials.The correlation between biological clock and magnesium nutrition and the MGTs family of magnesium ion transport gene was studied to explore the molecular mechanism of the regulation of biological clock and magnesium ion mutual feed in Arabidopsis thaliana.The main findings are as follows:1.Under normal magnesium ion concentration,the expression of MGT2 and MGT10 in the magnesium ion transport gene family MGTs of Arabidopsis has a circadian rhythm.2.The circadian rhythm of MGT2 gene expression in Arabidopsis is affected by extracellular low magnesium and high magnesium,while the biological rhythm of MGT10 gene expression is not affected by extracellular low magnesium and high magnesium.3.The change of extracellular magnesium ion concentration affects the circadian rhythm of clock core oscillator genes CCA1,LHY and TOC1in Arabidopsis circadian.4.Alterations in the expression of the circadian clock core oscillator genes CCA1,LHY,and TOC1 of Arabidopsis cause changes in the biorhythm of MGT2 and MGT10 expression.In summary,this study found for the first time that the change of extracellular magnesium ion concentration in plants affects the circadian rhythm of magnesium ion transport gene MGT2 and MGT10 expression and the circadian rhythm of circadian clock core genes CCA1,LHY and TOC1 expression,while the circadian clock regulates magnesium in the export pathway.The expression of the magnesium ion transport genes MGT2 and MGT10 suggests that plants may maintain their normal growth and development through the mutual regulation of magnesium nutrition and circadian core genes.The results of this study will provide a theoretical basis for the study of the molecular mechanism of plant circadian clock and magnesium ion feedback control,and provide a theoretical basis for the efficient use of crop nutrients in practice.
