Magnetic resonance(MRI)is an important means to study the properties and structures of matter.The traditional magnetic resonance technology is limited by its detection method,and can only be used for the detection of spin ensemble.The sensitivity and spatial resolution are not high,so it can not meet the needs of microscale material science research.A new magnetic detection technology based on NV color center in diamond has the advantages of high resolution and high sensitivity,and is expected to realize single molecule or nanometer scale detection.In order to use NV color center to realize the above microscopic magnetic resonance detection,the preparation of effectively controllable samples on diamond surface is a prerequisite,and the existing methods are to place molecules randomly or through average density control in diamond.Surface.DNA origami is a kind of complex nanostructure based on DNA base complementary pairing.through the exquisite design of DNA structure,DNA origami with various target molecules can be made.Because we can design DNA origami with target molecules only in a specific position,and various spin groups can be marked on the target molecules as needed,DNA origami is very suitable for the preparation of single molecule samples.It is used for microscopic magnetic resonance research based on NV color center.According to the needs of the study,we can arrange different nano-scale spin arrays on DNA origami,which is quantum computing.In 2005,quantum simulation and quantum precision measurement laid an important technical foundation.This paper mainly deals with the following work:(1)The design method and fabrication technology of DNA origami are explored,and a square structure DNA origami is made.(2)The process of attaching DNA origami to diamond surface was explored.The structure of square DNA origami has been improved many times,and a variety of modifications have been tried on the diamond surface.Finally,a series of suitable conditions have been found.The DNA origami has been successfully prepared on the diamond surface,which has good completion.And reached a very high density.(3)Two schemes for connecting specific proteins or DNA to DNA origami were designed,which can be characterized by atomic force microscope and fluorescence detection,respectively.We tried these schemes,fabricated and characterized DNA origami samples with single molecule or more complex molecular array,and obtained good experimental results.(4)A series of schemes were designed to bind arbitrary proteins to the square DNA origami we used.these schemes were tried and characterized by atomic force microscope,and then various experimental conditions were repeated and explored many times.
