Drought and water shortage have become a serious problem in the world's agricultural production.Due to the lack of precipitation and changes in precipitation patterns,drought and salt damage are the most destructive among many abiotic stresses,and it is also an important factor limiting crop quality and yield.broomcorn millet.One of the ancient gramineous food crops in China,it has the characteristics of short growth period,drought tolerance,thinness resistance,and rehydration ability after drought.It is an excellent material for studying the improvement of crop stress resistanceThe ABA-acid-stress-ripening(ASR)protein is a hydrophilic plant-specific protein that plays an indispensable role in abiotic stress in plants.Although ASR genes have been shown to respond to a variety of abiotic stresses in a variety of plant species and confer resistance to transformed plants.However,studies on the ASR gene family of broomcorn millet have not been reported.In this study,six broomcorn millet ASR family members were identified by comparing the broomcorn millet genome database,and by analyzing its gene structure,protein alignment and phylogenetic relationship and expression analysis of different tissues,it can be found that the C-terminal of Pm ASR protein contains highly conserved motifs.Phylogenetic analysis can divide the broomcorn millet ASR gene into three groups:(1)Pm ASR1,Pm ASR2,Pm ASR5(2)Pm ASR3(3)Pm ASR4,Pm ASR6.Subcellular localization analysis revealed that except for Pm ASR5 localized in the cytoplasm,the rest were in the nucleus.Through tissue expression analysis,the expression of Pm ASR1\ 2\ 3\ and 6 genes was root>stem>leaf,and the expression of Pm ASR4 and 5 genes was leaf>stem>root.The ASR family members with the highest expression in each broomcorn millet tissue were selected for cloning and bioinformatics analysis(root: Pm ASR1,stem: Pm ASR3,leaf: Pm ASR4)and the Pm ASR1,3,4 genes were analyzed by fluorescence in various tissues and The transcriptional profiles under different abiotic stresses do not contain signal peptides and transmembrane structures.The protein structure is mainly composed of ?-helix and irregular coil.The results show that the Pm ASR1 and Pm ASR4 genes are mainly The roots were up-regulated,and the Pm ASR3 gene was mainly up-regulated in leaves,and each gene was significantly differentially expressed under at least one stress or multiple stresses and both were highly up regulated under ABA hormone stress.Therefore,the ASR family gene of the broomcorn millet is an abiotic stress response gene.The results of this experiment not only facilitate the functional identification of the ASR family genes,but also lay the foundation for revealing the resistance of the broomcorn millet.
