Optical Clearing Method And Its Applications For Nude Mice Skin

Optical clearing technique is used to reducing the scattering coefficient of tissue by using optical clearing agent(OCA),which has high permeability to tissues and high refractive index,to help improve the depth of optical imaging.Nude mice was widely used in the field of Oncology,Immunology and other related medical biological research for its thymus having no T cell and hairless skin.Light transparency of the skin,as an important research field of optical clearing technique,has attracted a widely attentions,but it is still lack of research in nude mice skin,especially in vivo skin transparent.In order to solve the change of the microenvironment caused by the nude mice dorsal skin window chamber,we established a standardized optical clearing method to realize the optical clearing of nude mice dorsal skin.Moreover,the changes of optical clearing window chamber for nude mice dorsal skin were dynamic monitored,and we observed the recovery of the skin after experiment.Finally,successfully used skin optical clearing window chamber to observe vasoconstriction before and after vascular targeted photodynamic therapy.The skin optical clearing window of nude mice is expected to overcome the shortage of traditional skin window models of nude mice.In addition,the biological tissues has been observed without damaging the microcirculation.
