Purpose:To research the expression of H19 in skeletal muscle of rats in different exercise training modes and to analyze the correlation between physiological and biochemical indicators of skeletal muscle and H19,to explore whether the expression level of H19 can be used as a new indicator of exercise intensity(a marker of exercise intensity)and the role of H19 in the transformation process of skeletal muscle fiber type.Subject and Methods:1.Experimental subject: 96 SPF grade SD male rats,weighing 200-220 g.2.Experimental method:All of the rats were randomly divided into 4 groups and with 24 rats in each group.Group A: general aerobic exercise training group.Group F: chronic sports fatigue injury group.Group H: high intensity interval training group.Group C: control group without training.3.Experimental method:Rats in group A,F and H were trained on the animal treadmill for7 weeks,the principle refers to the animal treadmill model of Bedford et al.At week 1,3,5and 7,6 rats were randomly selected from each group of A,F,H and C to collect gastrocnemius muscle and blood.The morphological changes of the muscle during different exercise training were observed by HE staining.ATP staining was used to observe the transformation of type I and type II fibers in different training modes.The serum level of creatine kinase was measured by colorimetry,serum urea nitrogen levels were measured by diacetyl-oxime method,the serum succinic acid dehydrogenase level was determined by colorimetry,the serum lactate dehydrogenase level was determined by micro enzyme standard method,the serum malondialdehyde level was determined by microplate method.H19 mRNA expression was determined by quantitative real-time PCR.Results:1.The relatively expression of H19.The results of inter-group comparison of the same group at different training weeks showed that the expression of H19 in group A increased significantly at week 3.The expression of H19 decreased significantly in group H at week 5.The expression of H19 in group F decreased significantly at week 5.2.HE staining results.From week 1 to week 7,muscle fibers in group A were in neatarrangement,the nuclei were evenly distributed,and the cross-sectional area of muscle fibers showed an upward trend.From week 1 to week 7,muscle fibers in group H were orderly arranged and the nuclei were evenly distributed.The changes in the cross-sectional area of muscle fibers showed an upward trend and were more obvious than those in group A.From week 1 to week 3,muscle fibers in group F were more orderly arranged,the nuclei were more evenly distributed,and the cross-sectional area of muscle fibers was slightly increased.From week 5 to week 7,the arrangement of muscle fibers in group F began to be disordered,some muscle fibers were broken,and the nuclei were disordered and increased significantly.3.ATP staining results.The percentage of Type I fibers in group A increased at week 1,decreased at week 3,increased at week 5,and decreased at week 7.The percentage of Type I fibers in group H increased at week 1,3,5 and decreased at week 7.The percentage of Type I fibers in group F increased at week 1,3,5 and decreased at week 7.4.Results of serum biochemical indexes.The change trend of CK,BUN,SDH,MDA and LDH in group A gradually decreased from week 1 to week 3 and increased from week 5 to week 7.In groups H and F,CK,BUN,SDH,MDA and LDH increased gradually from week 1to week 5 and decreased from week 7.Conclusion:1.In this experiment,the general aerobic endurance training achieves the best training effect in the seventh week.After the seventh week,the training continues,and the indicators may tend to be stable.2.In this experimental scheme,HIIT training achieves the best training effect in the fifth week,after which the body begins to adapt to the intensity and starts to repair itself.3.In this experimental scheme,chronic fatigue injury training reached the strongest stimulation effect in the fifth week,and the maximum injury occurred.Then the body slowly ADAPTS to the intensity and starts to repair itself.4.HIIT training has the best effect by combining various physiological and biochemical indexes.5.H19 can be used as a new indicator of exercise intensity.6.H19 may play a important role in promoting the transformation of type II fibers.
