As an important desert species,Haloxylon ammodendron can adapt to drought,heat,salinity and other extreme environments,To study the expression regulation and resistance mechanism of resistance genes in Haloxylon ammodendron seedling,we can excavate the unique resistance gene resources,select excellent resistant materials and improve the process of desert ecological management,Which has a great significance to desertification control and ecological security.The sequencing and analysis of Haloxylon ammodendron drought and surface high temperature transcriptome were carried out in the early stage of the study group.Based on the previous study of the research group,six genes related to adversity were selected to learn the function of Haloxylon ammodendron NAC transcription factor,and then study the mechanism of its anti-stress.The main contents and conclusions are as follows:?1?HaNAC12 was cloned and expression patterns analysis of six NAC Transcription Factors from Haloxylon ammodendron.A NAC transcription factor gene named HaNAC12was cloned from Haloxylon ammodendron,The length of ORF was 912 bp and encoding 303amino acids,Sequences alignment showed that HaNAC12 contained five domains,which was conserved in NAC family.The real-time PCR analysis showed that expression patterns of HaNAC2,HaNAC3,HaNAC12,HaNAC20,HaNAC38 and HaNAC42 from Haloxylon ammodendron can be used to respond to stress,indicating that the function and working mechanism of the six genes are different under simulated drought,high salinity,simulated high temperature stress and IAA,ABA treatments.?2?Cloning and cis-acting element analysis of HaNAC3 promoter.The promoter of HaNAC3 was cloned from Haloxylon ammodendron seedling and the length was 1601 bp.The HaNAC2 promoter was cloned in the early stage of the study group.HaNAC2 and HaNAC3 promoters contain common cis-acting elements and contain different cis-acting elements.These different cis-acting elements may determine the differences in HaNAC2 and HaNAC3 gene expression patterns and functions.?3?The transgenic tobacco molecular detection and functional analysis of HaNAC2 and HaNAC3.The pCAMBIA1304-HaNAC2 and pCAMBIA1304-HaNAC3 plant expression vector were constructed and transferred into tobacco to obtain T1 transgenic tobacco.Under the simulated drought and high temperature treatments,the molecular detection of HaNAC2and HaNAC3 tobacco was carried out.The phenotypic and germination rates of transgenic tobacco were observed and analyzed under different treatments.Based on the analysis,HaNAC2 and HaNAC3 genes improved the resistance of tobacco to simulated drought and high temperature stress.
