Objective:With the further understanding of the pathogenesis of breast cancer,many signal pathways and therapeutic targets related to breast cancer have been discovered.In this study,we found that competing endogenous RNA,?ceRNA?network and SSP411 gene have an impact on the occurrence and development of breast cancer by means of bioinformatics and experimental verification.METHODS:1.The chip data were downloaded from The Cancer Genome Atlas?TCGA?database.The difference of Long non-coding RNA?lncRNA?was analyzed by R software,and then the ceRNA network was constructed and subjected to enrichment analysis of GO and KEGG.By considering the efficacy of each prognostic-related lncRNA,the prognostic-related lncRNA were fitted in multivariate Cox regression analysis,and the risk scoring model was constructed.2.Knockout of SSP411 vector were constructed.The SSP411 knockout vector was constructed.The constructed vector was transfected into breast cancer cell SKBR3 by cell transfection,and the stably expressed cell line SKBR3-SSP411-/-obtained by SSP411 knockdown was screened.The effects of SSP411 on breast cancer cell SKBR3 were investigated by Real-time PCR and Western Blot experiments based on gene expression results,protein expression results and cell validation results.Result:1.The results of this study show that five lncRNAs are associated with the prognosis of breast cancer.These five lncRNAs are not only at critical nodes in the ceRNA network,but also related to the overall survival of breast cancer patients.2.The SSP411 knockout SKBR3 cell line was successfully constructed and verified by Real-time PCR and Western Blot.The results of cell experiments showed that knocks out the SSP411 gene,the cell proliferation ability is decreased,the cell migration ability is decreased,and the cell cycle is blocked.Conclusions:1.There are five lncRNAs that may have potential for becoming prognostic markers of breast cancer.2.When the breast cancer cell SKBR3 knocks out the SSP411 gene,the cell proliferation ability is decreased,the cell migration ability is decreased,and the cell cycle is blocked.This indicates that 5 lncRNAs and SSP411 have potential as therapeutic targets for breast cancer.
