In recent years,with the rapid development of optoelectronic technology,optical interferometry has been widely used in ultra-precision machining,national defense,military industry,life sciences and other fields because of its advantages of high precision,high sensitivity,real-time,full-field measurement,non-destructive.In the mechanical experiment,it is very important to perform interference detection on the measured object,obtain the interference fringe pattern,and analyze the variation of the interference fringe to solve the actual problem.In this paper,several applications are analyzed in detail based on optical interferometry.The main contents of this paper are as follows:1.The principle of optical interferometry and phase-shift interferometry are introduced.The gray-scale signal and phase-type signal of phase extraction algorithm of time series are analyzed and discussed.2.A real-time monitoring system for crystal growth based on Mach-Zehnder interferometer is introduced.The observation principle and many functions of image analysis and processing software are analyzed.At the same time,the crystal growth liquid pool with artificial groove is introduced.3.An analytical method for solution concentration change during protein growth based on interference was proposed.Calculate the solution concentration curve by combining the phase correction method and the relationship between phase change and concentration change.The phase curve of the experimental group and the control solution are subtracted to obtain a protein phase curve.The dynamic relationship between water evaporation and solute adsorption was discussed.The effect of single factor on protein crystallization was obtained by analyzing the rising phase curve of protein and eliminating the influence of vapor pressure difference.This provides a new method for studying the influence of precipitators and additives on protein crystallization process.4.A real-time off-plane displacement tracking algorithm based on displacement compensation is proposed.The principle of displacement compensation measurement and tracking algorithm are introduced in detail.Gabor template is used for matching with subset of interference fringe pattern before and after displacement,so corresponding gabor templates are obtained.The displacement can be obtained through the phase shift of these two templates.In the experimental system,the algorithm can measure the out-of-plane displacement with a measurement range of 210 micron,a measurement resolution of 20 nm and a time resolution of 200 ms.It also has a strong anti-speckle noise performance because it does not need complicated accurate phase calculation.
