In the field of three dimension,Hi-C technology play a key role in genome-wide interaction researching.As technology advances,more and more problem in traditional Hi-C has been revealed such as low data utilization ratio,long experimental cycle,high cost and so on.In order to make up these shortage,there is a new technology named “DLO Hi-C(Digestion-ligation-only Hi-C)” published on Nature Genetics in May of 2018.Compared with the traditional Hi-C,DLO Hi-C has many advantage like high sequencing quality,short experimental cycle,good repeatability and simple operation etc.Most of analyze tools for traditional Hi-C can’t apply to DLO Hi-C because of the different experimental operation,so it’s very important to develop a tool which can analyze DLO Hi-C data.We develop a new analyze tool named “DLO Hi-C Tool”,DLO Hi-C Tool integrate the partial function of ChIA-PET Tool and MAFFT and traditional Hi-C protocol,then construct a integrative DLO Hi-C analyze tool by Java.DLO Hi-C Tool using easily,needn’t manual intervention in all process,less dependence and running fast.In addition to analysis of one-click,DLO Hi-C Tool also support distribution operation and provide some script of file conversion and statistic to satisfy diverse need of users.If user’s server support graphical user interface,DLO Hi-C Tool can generate a GUI and make parameter setting and survey the progress of analysis more visible.The test data we used is coming from the article of DLO Hi-C,and we show the partial function and analyze result of DLO Hi-C Tool.We test 6 group of DLO Hi-C data,5 of this group is in-situ DLO Hi-C.We use same parameters for all test data and the running time of different data is between 3 and 7 hours.We find out that the result of DLO Hi-C Tool was similar to the result of articles,and discovered more TAD structures in THP1 cell line.
