Anopheles sinensis is the most common mosquito species in China,widely distributed in most provinces in China.It is not only an important medium for the spread of malaria,but also can spread other diseases such as filariasis.It is important to study the transcriptome of Anopheles sinensis in order to understand the molecular level of the host and the mechanism of olfactory detection,and to study the olfactory sensor.In this paper,the total RNA was extracted from the antennae of female Anopheles sinensis before and after blood-meal,and the genes related to olfaction were extracted according to the dominant role of the blood preference and its sense of smell in the reproduction of female mosquitoes.Odorant binding protein,olfactory receptor protein and ionotropic receptor protein gene,which are closely related to olfaction,to study the changes of these three kinds of olfactory related genes in order to find out the key genes of olfactory function and its relationship with locating host behavior.An Anopheles sinensis feeding platform was set up and the Anopheles sinensis was extensively cultivated.In this paper,the use of mosquito body at the beginning of the analysis of detection,before and after the blood-meal transcriptome data no significant difference,as the olfactory sensory focus on only a slight weight due to antennae.Therefore,this research changed to a detailed,hard work,for each female mosquitoes to pick up a pair of antenna to experiment,and ultimately obtained satisfactory results.A total of 10,570 new transcripts were predicted and analyzed in the antennal transcriptome of Anopheles sinensis before and after blood-meal.And the gene structure of 8288 genes in the original genomic annotation was optimized.Finally,gene expression level and differential expression analysis of odorant binding protein gene,olfactory receptor protein gene and ionotropic receptor gene were carried out.The results showed that there are 42 odorant-binding protein genes,45 olfactory receptor genes and 48 ionotropic receptor genes which are expressed in the antennal transcriptome.5,3 and 2 are new discovery.16,5 and 5 are significantly enhanced.5,2 and 2 are significantly decreased.These olfactory-related genes with reduced expression in female mosquitoes are likely to have a close internal relationship with their locating host behavior.In this paper,the sequence analysis of Anopheles sinensis transcriptome and the simultaneous analysis of the three olfactory-related genes in mosquitoes were not reported in previous literature.The results of this study were to identify the olfactory key genes related to the host Which laid a foundation for the further study of the related odor receptor function.
