On the basis that it's found in the early experiment that expression of CdMYB59 is related to dwarfing and drought resisting of cynodon dactylon in a certain way,this research intends to analyze functions of transgene rice via transgene rice which overexpresses CdMYB59 and establish genetic transformation system of cynodon dactylon to lay the foundation for further obtaining transgene cynodon dactylon.The major experimental results are as follows:1.Obtaining and phenotypic analysis of transgene rice plant that overexpresses CdMYB59We obtained transgene rice plant that overexpresses CdMYB59 by agrobacterium-mediated transformation;10 from them were identified via PCR detection as positive transgene rice plants and their homozygotes were obtained.Detected by real time fluorescent quantitative PCR and expressions of CdMYB59 in all the 10 transgene rice plants were found.Carried out phenotypic observations of 14-day homozygote plants of T2 transgene rice and wild plants and found No.O1-3,O4-10,O7-17,O9-4 and O10-17 homozygote plants were higher than wild plants;while No.O2-5,O3-8,O5-1,O6-13 and O8-7 were lower than wild plants.We also measured root lengths,and the result shows that root lengths of strains of transgene rice plants were 48%-106% longer than those of wild rice plants which were separated from T2 transgene rice plants.We also analyzed drought resisting capacities of transgene plants.It's found in the results of T2 transgene rice plants via osmotic stress(PEG-6000)and wild rice plants that relative water contents of transgene plants were obviously higher than those of wild plants,36%-75% higher;and relative conductivities of transgene plants were clearly lower than those of wild plants,only 42%-55% of wild plants.The result shows that expression of CdMYB59 is able to improve rice plants' drought resisting capacity.2.Established genetic transformation system of cynodon dactylon and obtained transgene cynodon dactylon that overexpresses Cd MYB59We further established a kind of genetic transformation method mediated by Agrobacterium on the basis of stable and highly efficient regeneration system of cynodon dactylon.We got 8 regenerated plants of resistant calluses by this method and detected by PCR,phenotype of 2 of the transgene regenerated plants was positive.
