High frequency(kHz)biphasic electrical stimulation can block the conduction of action potential on nerve, and have contribute to many disease treatment, such as alleviate chronic pain, relieve muscle spasm and improve urinary efficiency etc.So applying high frequency electrical stimulation in clinic safely and efficiently would have a great value. However this technique would introduce injuries for nerve while achieving blocking effect which becomes a key problem to reduce and even eliminate the injuries when accomplishing conduction block. This study designed the biphasic asymmetric waveform and analyzed the effects of high frequency electrical stimulation with different parameters and laser irradiation on blocking conduction on nerves through the variation of CAP's amplitude based on the data gathered from isolated bullfrog's sciatic nerve. And giving a more safe and efficient method by the results of experiments in high frequency electrical stimulation.First, this study used biphasic asymmetric discontinuous square wave of high frequency electrical stimulation to make sure the net charge was zero, under this premise, studied the blocking effects and injury effects of high frequency stimulus with different frequencies(F=1,2,5 kHz)and different anode-to-cathode pulse width ratios(r=1,2,3).The experiment results showed that high frequency electrical stimulation with lower frequency and bigger anode-to-cathode pulse width ratio could get a better blocking result;high frequency electrical stimulation with higher frequency and smaller anode-to-cathode pulse width ratio could get a better injury result;overall consideration of the effects of blocking and injuring, the high frequency stimulation with the parameter combination r=1,F=5 kHz;r=2,F=5 kHz;r=2,F=2 kHz could get a better result. In order to reduce the injury introduced by high frequency electrical stimulation, this study investigated the effects of laser irradiation with different frequencies(1 kHz,2 kHz,2.5 kHz,4 kHz,5 kHz,10 kHz)on accelerating the recovery for injured nerve. The result suggested 650 nm and 25 mW laser irradiation could reduce the injury caused by high frequency electrical stimulation and promote the recovery speed for injured nerve. For the study of laser irradiation with different frequencies selected in this study, the experiment results indicated that laser irradiation had a timeliness in the aspect of reducing the injury introduced by high frequency stimulus. And the effects of 5 kHz laser irradiation could get the best results,2 kHz and 2.5 kHz laser irradiation could get a relatively superior result. For the other frequency(1 kHz,4 kHz,10 kHz)laser irradiation, there were some inhibiting effects for the recovery of injured nerve, and especially 10 kHz was the most significant. This result indicated that laser irradiation with appropriate parameter could be used as a supplementary mean to reduce the injury caused by high frequency electrical stimulation and accelerate the recovery of injured nerves. The result of this study indicated that select the stimulation waveform with appropriate parameters and use laser irradiation could reduce the injuries caused by high frequency stimulation, which could provide a theoretical guide for the application of high frequency electrical stimulation in clinic.
