Coastal wetland is one of the most important life support systems on the earth.It is a place for many birds to live,migrate,wintering and reproduce.It is also an excellent living place and ideal habitat for biogas and aquatic organisms.Scirpus mariqueter is endemic to coastal wetland of Yangtze River Estuary and Chinese pioneer plant,has ecological service function of reduction wave,soil improvement,carbon sequestration and purification,but also a good habitat for wild animals.However,since 1990 s,with the rapid development of coastal economy,increasing the reclamation of coastal wetland resulted in large area shrinking rapidly,Scirpus mariqueter wetland landscape area is decreasing.At present,most researches on vegetation restoration and reconstruction of Scirpus mariqueter have been focused on seed reconstruction and tidal action,but few of soil microbes have been studied.Therefore,the protection of Scirpus mariqueter community and restoration work is extremely important.In this paper,through the research of the Biolog micro plate method and high-throughput sequencing technology on different regional of Scirpus mariqueter Rihizosphere soil microorganisms,combined with soil physicochemical factors,in order to find the effects of soil microbial community structure,so as to find out the influence reason of Scirpus mariqueter distribution,provide the theoretical basis for establishing a restoration of the ecological community plan.The main results are as follows:(1)Through the analysis of physical and chemical factors shows that : the difference of tidal flat elevation and water content and salinity are major factors of Scirpusmariqueter distribution pattern in Nanhui Dongtan wetland.The height of the study area is between 3.3-4.7m,which is high in the middle and low in both sides in 2015,With the deepening of siltation,in 2017,the elevation decreased from side to sea and rasied on both sides of the East and West.In 2015 the beginning of the study,Scirpus mariqueter community mainly concentrated in the Middle East study area,namely higher sedimentation zone,with the deepening,the Middle East region is gradually heightening,Scirpus mariqueter community westward migration.The Scirpus mariqueter disappeared in eastern zone,is to replace the Spartina alterniflora.The soil pH is 8.679,alkaline,Scirpusmariquetergrow in alkaline soil.(2)By Biolog micro plate method to analyze the relation between the functional diversity and environmental factors in the different regions and the rate of single carbon source,the rhizosphere soil microbial community utilization ability of single carbon source: dense area >scarce area> light beach.The main factors affecting the rhizosphere microbial activity is soil salinity,particle size and moisture content.Environmental factors and soil microbes interact each other.(3)By Misqe sequencing in different regions of soil microorganisms season analysis:Samples of microbial structure similarity,there are 15 kinds of common bacterial phyla,including Proteobacteria?Bacteroidetes?Acidobacteria?Chloroflexi?Planctomycetes?Actinobacteria?Cyanobacteria?Gemmatimonadetes are the dominate bacterias.The change of Cyanobacteria in the different samples are greatly.The diversity and abundance of microbial communities varied significantly in seasons.The diversity and abundance of microbial communities in the same sample were lower in spring and summer,and higher in autumn and winter.In the same season,soil microbial community diversity and abundance: dense area > scarce area> light beach.Soil salinity,water content and nitrate nitrogen are the main factors affecting the diversity of microbial community.The artificial intervention sedimentation tidal should be take in the future,optimize of Scirpus mariqueter soil conditions,quantify the soil micro mechanism in the restoration of Scirpus mariqueter is the next important direction of the study.
