Objective:Personality describes the steady individual differences of mental state in cognition,emotion and motivation which leads to the steady behavior inclination.About 2/3 personality traits differences are due to the impact of gene.Therefore people are trying to figure out the gene that can influence personality traits through gene polymorphrism research.5-HT and dopamine are associated with personality traits and any other substances that can have an impact on dopamine or 5-HT are also associated with personality trait and this kind of association differs in gender.The activation of ADORA2A can affect many receptors,it can play a relative and inhibition role in dopamine receptors.ADORA2A itself can influence learning memory,habit cultivation and the lack of ADORA2A can lead to behavior excitement,aggressivenessincreaseandgoal-leadingbehaviorenforcement.Furthermore ADORA2A has been proved of having relationship with several kinds of neural system diseases.So we can speculate ADORA2A gene may be related with personality traits.Since there hasn't been any relative research in China,we decide to use the method of PCR-RFLP to detect ADORA2A gene polymorphism which aims to discuss whether there is a relationship between ADORA2A gene polymorphism and personality traits in healthy Chinese Han subjects.Method:Our study choose 535 participants in total which have no mental disorder or other serious physical diseases history,including 259 males and 276 females.First,we use TPQ personality scales to assess scores and collect 2ml peripheral vein blood respectively at the same day.Second,we extract DNA from the blood through KI method.Third,we detect their gene types through PCR-RFLP.Finally,we conduct statistic analysis through SPSS software to detect whether there are differences in the scores of different gene type groups,P<0.05 is seen as significant statistical significance.Result:1.Among the total of healthy Han population,for rs5996696,NS(X~2=1.366,P=0.505),HA(X~2=1.143,P=0.565)and RD(X~2=2.310,P=0.315)scales have no significant differences in the scores of different gene type groups.For rs2298383,NS(X~2=3.101,P=0.212),HA(X~2=4.161,P=0.125),RD(X~2=0.748,P=0.688)scales also have no significant differences in the scores of different gene type groups,but HA4(X~2=6.134,P=0.047)sub-scale has significant differences.2.In gender stratification,for rs5996696,in males NS(X~2=4.353,P=0.113),HA(X~2=1.152,P=0.562)and RD(X~2=2.303,P=0.316)scales have no significant differences in the scores of different males gene type groups,but RD4(X~2=8.348,P=0.015)sub-scale has significant differences,in females NS(X~2=0.06,P=0.970)and HA(X~2=1.272,P=0.529)scales have no significant differences in the scores of different females gene type groups,but RD(X~2=6.135,P=0.047)scale has significant differences.For rs2298383,in males NS(X~2=2.417,P=0.299),HA(X~2=0.971,P=0.615)and RD(X~2=4.907,P=0.086)scales have no significant differences in the scores of different males gene type groups,but NS3(X~2=7.793,P=0.020)sub-scale has significant differences,in females NS(X~2=2.473,P=0.290),HA(X~2=4.604,P=0.100)and RD(X~2=0.463,P=0.793)scales have no significant differences in the scores of different females gene type groups,but HA4(X~2=6.210,P=0.045)sub-scale has significant differences.Conclusion:ADORA2A gene polymorphism has relationship with personality traits in healthy Chinese Han subjects.ADORA2A gene may be a potential gene that can influence personality.
