Objective: To study the effects of ethanol on the lifespan and its related indicators in model organism Caenorhabditis elegans,and explore the mechanism preliminarily.Methods:To set the ethanol groups(530mmol/L ethanol,53mmol/L ethanol,5.3mmol/L ethanol)and the blank group.Through the ordinary life test,reproductive test,pharynx-pumping,body-swing rate measurement,heat stress life test and daf-16 mutant strains nematodes life test to analyze and evaluate the influence of ethanol on lifespan and its related indicators of nematodes life.The mechanism of extending the life span of nematodes was preliminarily explored.Results:(1)The ordinary lifespan test: Ethanol groups(530mmol/L ethanol,53mmol/L ethanol,5.3mmol/L ethanol)compared to blank control group,the average lifespan of elegans in the ethanol groups was prolonged significantly(P < 0.01);(2)The reproductive capacity test: ethanol groups(530mmol/L ethanol,53mmol/L ethanol,5.3mmol/L ethanol)compared to blank control group,the reproductive capacity of each ethanol groups was not statistically significant different(P>0.05);(3)The pharynxpumping rate measurement: ethanol groups compared to blank control group,the pharynx-pumping rate of each ethanol groups was not statistically significant different(P>0.05);(4)The body-swing rate measurement: ethanol groups compared to blank control group,the body-swing rate of each ethanol groups was not statistically significant different(P>0.05);(5)The heat stress test: Ethanol groups(530mmol/L ethanol,53mmol/L ethanol,5.3mmol/L ethanol)compared to blank control group,average lifespan of elegans in ethanol groups was prolonged significantly(P < 0.01);(6)The Daf-16 mutant strains elegans life test: daf-16 mutant strains(530mmol/L ethanol)group elegans of the average life span was shorter than that blank group,but it was not statistically significant(P > 0.05).Conclusion: The appropriate concentration of ethanol can prolong the life span of the elegans,while it has not significantly affected its fertility and health at the same time.The appropriate concentration of ethanol can delay the aging of nematode worms to prolong their life span by the role of the IIS signaling pathway in the DAF-16 or increasing their anti-stress ability.
