This article mainly by wuwei liangzhou district crown pear planting as the research object,using different extraction parameters for optimization of the crown pear polyphenols extraction,and compare the three different extraction technology is used to crown pear polyphenols extraction effect,effect evaluation is extracted by extraction rate as index,find out the best extraction process parameters of the crown pome,finally study the best extraction process of polyphenols in the crown pear and crown pear polyphenols extraction and DPPH · · OH radical scavenging ability of determination,using ultraviolet-visible spectrophotometry to determine the effect of free radical clearance,to judge from the crown pear polyphenols antioxidant activity.The results are as follows:1.Single factor orthogonal experiment was used to explore the optimal process parameters for extracting polyphenols in crown pear from organic solvent.The experimental results show that the main factors of the four factors affecting the extraction of polyphenols from the organic solvent are extraction temperature,extraction time,liquid ratio and solvent concentration.So using organic solvent to extract the crown pear polyphenols in the optimum process parameters for the 35:1 plead to 60% ethanol,in 65 ? water bath to crown pear polyphenols extraction of 50 min,extracting 3 times,under this condition,calculate the polyphenols extraction yield of 16.286 mg/g.2.Single factor test and orthogonal experiment design were used to study the optimal extraction process parameters of microwave assisted extraction of polyphenols.The experimental results show that the main factors of the four factors that influence the microwave assisted extraction of polyphenols in crown pear are: the concentration of organic solvent,the ratio of the material liquid,the microwave power and the extraction time.Microwave assisted extraction of polyphenols from crown pear is the best process parameters: with 40:1 40% ethanol,under the condition of setting up 450 w microwave power and extraction of 100 s,extracting 3 times,in this condition,the polyphenol extraction yield of 16.114 mg/g.3.Single factor test and orthogonal experiment design were used to explore the optimal process parameters for extracting polyphenol from crown pear by ultrasonic assisted method.Experimental results show that the influence of ultrasonic extraction crown pear polyphenols for primary and secondary factors: extractant concentration > solid-liquid ratio > extraction time > extraction temperature,therefore,the best extraction technology conditions for polyphenols: i.e.,according to the material liquid than 50% ethanol for 40:1,at 30 ?,60 min ultrasonic wave extraction,extraction of 3 times,under this condition,crown pear polyphenols extraction yield of 16.369 mg/g.4.Through the organic solvent and microwave assisted comparing with the results of ultrasonic assisted the extraction of organic solvent method,shows that ultrasonic assisted organic solvent to extract the crown pear polyphenols,the highest extraction rate,increased from 16.114 mg/g to 16.369 mg/g,then achieve the cost savings,to enhance the efficiency of the production.5.Using the Vc standard as the control,the removal ability of crown pear polyphenols on DPPH free radicals,OH free radicals and O2-free radicals was determined by visible spectrophotometry.The results showed that crown pear polyphenol DPPH and ·OH free radicals and O2-free radicals both had certain clearance ability.In particular,the scavenging ability of hydroxyl radical is better than Vc,and the removal ability and the content have a certain amount of relationship,the higher the concentration,the stronger the clearance ability.
