In order to satisfy people's need for early diagnosis and treatment of diseases,achieve the low detection limits and high sensitivity of molecular diagnostics,a variety of techniques are being utilized.The novel molecular diagnosis proposed in this paper is convenient and quick,combining the complex nanoparticles containing vitamin C fragments with methylene blue(MeB).According to the nanoparticles' characteristics such as large specific surface area,we realize the accumulation of polyvitamin C in the detection system.Take full use of the reduction property,vitimin C reacts with methylene blue in signal amplification,our purpose is to achieve qualitative and quantitative detection of disease markers.The specific work as follows:1.Preparation of polymer materials:Polymer materials which have polyvitimin C and PEG as hydrophilic units,PS as hydrophobic unit,are synthesized according to the RAFT.Their chemical structure is characterized by NMR and GPC.The PDI calculated by GPC is 1.4,which means the size distribution is narrow,the polymerization is controlled.2.Preparation of complex nanoparticles:The condition of co-self-assembly nanoparticles were optimized by DLS in order to produce homogeneous size nanoparticle.The morphology and stability of nanoparticles is examined using TEM and Fluorophotometer.Complex NP has core-shell structure with a diameter of 160 nm.3.Establish a new system of signal amplification:Vitamin C reacts with hydrogen peroxide producing hydroxyl radical which can make the color change of methylene blue from blue to white.It is easy to be observed.Meanwhile,UV spectrophotometer(UV)is used to optimize the conditions of the system.Finally the system can be used to achieve qualitative and quantitative detection of disease markers.Finally the concentration of nanoparticles is equal to methylene blue.There is 6%of H2O2 in the system.It can be examined after 2 minutes.4.Qualitative and quantitative detection of protein:Preparation of nanomagnetic beads(MB).The structure and morphology are characterized by FT-IR and TEM.Combining the new system of signal amplification with immunomagnetic beads,the detection is determined by double sandwich method,monitored by UV.It improves the qualitative and quantitative detection of disease markers,enabling the detection to be carried out from the traditional plate surface to the solution.In addition,the addition of nanomagnetic beads can make up for the lack of detection limits of amplification system.The detection linear range is 600-2000 ng/mL,with LOD is 400 ng/mL.
