Hypochlorous acid(HCl O)and hydrogen peroxide(H2O2)are important components of ROS and are considered to be potent antibacterial agents in life to kill bacteria during the immune response of organisms.Cysteine(Cys)plays an important role in maintaining the transduction and redox homeostasis of a variety of reactive species.Hypochlorous acid is an active species in tumor cells,The current treatment methods for tumors include surgical resection,radiotherapy,liver transplantation,and other clinical treatments.Chemotherapy and drugs are the mainstay of conventional treatment,but they lack targeting.At this stage,we are looking for a scientific method that can accurately locate these tumor cells and can be applied to the clinic.Therefore,this paper designed fluorescent probes with targeted recognition of tumor cells.It is targeted by indomethacin screening and hypochlorous acid expression.It is capable of bringing probe-targeted recognition into cells and releasing fluorophores.The probe was found to be able to distinguish normal cells from tumor cells through cell imaging.Among them,the imaging effect of 9L cells in rat glioma cells is the best.In addition,the synthesis of fluorescent probes based on H2O2and Cys was also designed.In this paper,methylene blue,a near-infrared fluorophore,is mainly used to design and synthesize targeted near-infrared fluorescent probes.(1)Near-infrared fluorescent probes IMC-1a and R-1,one end of the linking group ethylenediamine is connected to the near-infrared fluorophore methylene blue,and the other end is connected to indomethacin(IMC)and the reference substance p-methylphenylacetic acid,respectively.and the photophysical properties show that the probe IMC-1a can respond quickly(30s)and has good stability.Finally,the probe was successfully applied to rat glioma cells(C6,RG2,9L),human non-small cell lung cancer(A549),and human cervical cancer cells(He La)cells.imaging.(2)Using a long-chain linking group 1,2-bis(2-aminoethoxy)-ethane to connect one end of methylene blue and the other end to indomethacin(IMC)and the reference substance p-methylphenylacetic acid.Near-infrared fluorescent probes IMC-2a and R-2.The photophysical properties show that the probe IMC-2a can respond quickly(30s)and has good stability.Finally,the probe was successfully applied to the imaging of cells in rat glioma cells(C6,RG2,9L),human non-small cell lung cancer(A549),and human cervical cancer cells(He La)and after sacrifice by tail vein injection.Imaging of mouse adjacent tissue cells can be used for hypochlorous acid detection and cellular bioimaging.(3)Connect 4-bromo-1,8-naphthalenedicarboxylic acid anhydride through double diboron,then connect N-tert-butoxycarbonyl-1,2-ethylenediamine and coumarin,and finally react with acryloyl chloride to design and synthesize the Fluorescent probe F1 for the detection of H2O2and Cys.The photophysical properties show that the probe has good selectivity and stability to H2O2and Cys.
