Sequencing Complete Mitochondrial Genome Of Seven Murina Species(Chiroptera:Vespertilionidae) From China And Studying On Their Taxonomic Status

Abstract/Summary:
Known from Asia and Australasia,the genus Murina is a taxon of forest dependent bats,whose highest diversity is found in the tropical forests.Relatively rare in systematic collections and seldom abundant in the field,there is disagreement over the taxonomic status of certain closely related species.The genus has a rather uniform appearance.Externally,differences are primarily those of size,colour and ear morphology are similar.And cranial is similar,too.In addition,the genus also have sexal dimorphism.So,it is very difficult to identify the species only on the traditional morphological characteristics.Since DNA barcoding of the mitochondrial gene cytochrome c oxidase submit I(COI)has been a useful taxonomic tool in combination with traditional morphology for species identification and discovery.The discovery and description of new species in Murina have been published quickly.However,the phylogeny of the genus has not yet been known,and the research developed still slowly.In this study,specimens were captured in the wild and identified by traditional morphology and phylogenetic analysis based on COI genes,Cyt b genes and ND1 genes.After ensuring that their species were identified correctly,their mitochondrial genome was amplified and sequenced and annotated.By comparing the phylogenetic tree based on one gene,we would like to know the phylogenetic ability of different genes and what the question can be soulved by different markers.According to the mitochondrial genome can be the “Link” to connect the other maker in Genbank-nt.The results are as follows:(1)We get the total mitochondrial genome of seven Murina.M.harrisoni is16601 bp,M.eleryi is 16449 bp,M.lorelieae is 16430 bp,M.fanjingshanenesis is16488 bp,M.cyclotis is16442 bp,M.chrysoaetes is 16335 bp,M.shuipuensis is16584 bp.The mitochondrial genome of seven Murina was compared with the published data in NCBI-nt: the composition and structure of all mitochondrial genomeare the same,including 13 protein encoding genes,22 t RNA genes,2 rRNA genes and a control regions(D-loop).The anti-codon of mitochondrial genome tRNA of ten Murina is the same.There are few differences in the strat-codon and the stop-codon.(2)The phylogenety trees reconstructed from the nucleclotide alignment of COI gene,Cyt b gene and ND1 gene were compared.The phylogenety of different genes are different,which because of the selection pressure of different genes,or the difference of the rate of gene evolution.In addition,one gene could not give the correct phylogenety of the Murina because of the bad support nodes,and multi gene analysis was the only way to restore the early species divergence.Finally,we suggeste use the COI gene as the DNA Barcoding for identify species.The phylogenety of ND1 gene is similar to mitochondrial genome.(3)Mitochondrial genome can give more accurately information of the early phylogenety,and can assist in the revision or determination of traditional classification systems.Cyclotis-group and suilla-group,identification based on tooth characteristics,have not been confirmed by the mitochondrial genome,which can be attributed to the repeated adaptation of different branches to the approximate habitats,that is,the convergence evolution.And the phylogenety of Cyt b and COI gene,the phylogeny of the mitochondrial genome shows the relationship between the subfamily,and the subfamily of the Murininae is divided into Harpiocephalus,Murina and Harpiola,and the bootstrap values is high.(4)Mitochondrial genome can be used as a "joint" tool to link different(or section)marker sequences in databases.The different genes of Murina in the NCBI database do not include all speices.The mitochondrial genome can play a tool to link the sequences of different markers in the mtDNA,and the information can be found in time to identify the published sequence of species identification error,so that the molecular identification of the Murina can be easily identified.In this study,we used mitochondrial genome to sort out some erroneously sequences in NCBI-nt database.
