Objectives:Resveratrol and adipose derived mesenchymal stem cells have anti-aging effects.This paper aims to compare the two combinations with the anti-aging effect alone,and explore the mechanism.Methods:Human dipose mesenchymal stem cells(hADSCs)and human Dermal fibroblasts(HDFs)were isolated from human eyelid subcutaneous adipose mesenchymal stem cells(dipose mesenchymal stem cells hADSCs)by enzyme digestion-tissue mass culture.The phenotypes of hADSCs stem cells were identified by flow cytometry and induced by lipogenesis,identification of type ? and ? collagen in HDFs by immunohistochemical staining.The natural aging cell model of HDFs was established.Resveratrol,human adipose mesenchymal stem cells(hADSCs)and self were cultured in transwell co-culture chamber.However,the aging fibroblasts(HDFs)were co-cultured in groups.The cells were divided into four groups:(1)hADSCs acting on senescent fibroblasts group,(2)RSV alone acting on senescent fibroblasts group,(3)hADSCs RSV synergistic action group,(4)control group.Protein imprinting method was used to detect SIRT1 and ?-catenin protein of the cells in each group after co-culture.The expression of COL1A1,COL3A1,MMP-1 and GLB1 in fibroblasts was detected by qRT-PCR.Results:Cells extracted from human eyelid subcutaneous fat,the phenotypes of hADSCs stem cells were identified by flow cytometry: CD90,CD105,not CD34,CD45.The results were in line with the specific expression of mesenchymal stem cells.Cells extracted from human eyelid dermis,immunohistochemical staining expression type?and ? collagen,the identification results were in accordance with HDFs.After two weeks of culture,the cells were cultured in lipopolymers.The formation of lipid droplets was observed in transwell.After co-culture,The expression of SIRT1 and ?-catenin in co-cultured cells was detected by protein imprinting method.The expression of SIRT1 and ?-catenin in hADSCs+RSV group was higher than others.The expression of COL1A1,COL3A1 was detected by qRT-PCR.The tendency of it expression in hADSCs+RSV group was higher than others.The expression trend of MMP-1 and GLB1 was lower in hADSCs+ RSV group than in hADSCs,RSV synergistic group.Conclusion:Primary culture derived from dermal fibroblasts after natural senescence,the anti-aging effect of RSV combined with hADSCs was better than that two alone,and was related to mesenchymal epithelium transformation(MET).
