Transcription factor is a protein that binds to a specific nucleotide sequence upstream of a gene,which regulates the transcription of its gene.Transcription factors can regulate the binding of ribonucleic acid polymerase(RNA polymerase,or RNA synthetase)to DNA template.Transcription factors generally have different functional regions,such as DNA binding domains and effect domains.Transcription factors not only with the upstream promoter region of the gene binding,but also with other transcription factors to form a transcription factor complex to affect the gene transcription.WRKY family transcription factor is a plant-specific transcription factor,which plays an important role in plant metabolism and stress response.WRKY protein is conserved due to conserved WRKY domain of N-terminal 60 amino acids.Due to the rich structure of WRKY protein,But also a variety of,in the current study of WRKY protein,mainly for its non-biological and biological stress and long-development process of the regulatory role.The main result of this thesis are as follow:(1)Bioinformatics analysis of OsWRKY97,which encodes 233 proteins,and its promoter sequence contains a number of cis-acting elements associated with stress induction.Quantitative qRT-PCR was used to study the effect of rice In the expression pattern,found that the expression of tissue specificity and can be induced by different stress,are more adversity induced genes.(2)D-163 + 1300-Os WRKY9 7-GFP vector was constructed by D-163 +1300-OsWRKY97 overexpressing vector,D-163 + 1300-OsWRKY97 reverse expression vector,transgenic rice mediated by Agrobacterium tumefaciens,18 strains of transgenic plants with TO generation were obtained,and 23 strains were shown to inhibit the expression of transgenic plants.(3)The OsWRKY97 transgenic plants of T1 generation were obtained and the resistance of transgenic plants was tested.It was found that overexpressing plants enhanced the resistance to partial stress.
