MSAP method was used to explore the epigenetic diversity variation characteristics of Flaveria bidentis genome DNA.Analyze the difference geographical distribution regions of Flaveria bidentis populations,reveals regularities about different environment influence on epigenetic diversity variation of Flaveria bidentis.learn about different acquisition standards and different approach methods on genome DNA methylation patterns of Flaveria bidentis.providing theoretical foundation for Flaveria bidentis scientific gather strategy.The results showed that a total of 993 MSAP bands were amplified and the polymorphic loci percentage of which was 88.52%.The polymorphic loci percentage of Flaveria bidentis species in different invasion area is between 84.03%and 92.31%.GS,is 0.07and Nmis 3.321 which showed that there were frequent gene exchanges between Flaveria bidentis species and Genetic differentiation between populations was not significant,but there were significant differences in methylation patterns.We guess that the Flaveria bidentis may alter its methylation level to adapt to new environment.A total of 998 MSAP bands were amplified in seed germination experimentand polymorphic percentage was 95.29%.Changes of DNA methylation and demethylation can be found in 49%loci,and happened at the same time,of which there were 73 demethylated loci and 21 methylated loci,indicating that DNA methylation changes during seed germination mainly referred to the demethylated form,and the number of demethylated sites continued to rise rapidly in four days.The results was in consistent with the conclusion that seed germination needs lots of gene expressions.In organ and devemental stage test of the Flaveria bidentis,we got 536 and 407 MSAP bands respectively,and both of which have significant specificity.Individual differences between organs is discrepancy,the difference of root and stem is larger than leaf,and different development stages also have significant differences.Study on dynamic changes of Flaveria bidentis DNA methylation under different processing showed that a total of 340 MSAP bands were amplified,and the number of MSAP bands as well as methylation patternsis closer from the same primers.We thought that there was no influence on DNA methylation level of Flaveria bidentis towards different treatments.
