Sponge is a kind of filter-feeding multi-cell animal.With a special cavity structure,It becomes a kind of ideal host of microorganisms.The study found that sponge associated microorganisms can produce a wealth of active substances.It is a valuable topic to carry out the study of sponge associated microorganisms and their metabolites.It is of great significance to the study of new resources,novel compounds.This paper researches the diversity and inhibition activity of sponge Pseudoceratina sp.associated bacteria from Xieyang Island,Beihai City,Guangxi Details are as follows:1.First,the sponge Pseudoceratina sp.associated bacteria were isolated by six kinds of medium and purified with ISP2 medium.A total of 54 strains of bacteria were obtained.16S rRNA sequence alignments were performed and 24 strains of bacteria were gained after the repeated strains were excluded,They belonged to 3 phylum,4 classes,9 orders,13 families,14 genera.The dominant genus among them was Bacillus sp.,and a potential new strain BGMRC 2118 is also obtained.2.Polyphasic taxonomy of sponge Pseudoceratina sp.associated bacteria BGMRC 2118:The sequence similarity ratio of 16S rRNA between BGMRC 2118 strain and the closest available published strain Bacillus abyssalis SCSIO-15042T(JX232168)was 96.78%,which is less than 97%.It may be a new species or a new genus.And it was found that it remained stable homology with Bacillus sp.in N-J tree and M-L tree.Combined with the experimental results of its appearance characteristics,physiological and biochemical characteristics,numerical identification systems,chemical classification characteristics,and genotypic characteristics,it could be determined that BGMRC 2118 was a new species in Bacillus sp..3.Separately,The antibacterial activities of sponge Pseudoceratina sp.associated bacteria were studied using agar diffusion method,disk diffusion method,Oxford cup method.It was found that BGMRC 2216 and BGMRC 2127 could inhibit the activity of Penicillium marneffei and Vibrio parahaemolyticus,However,BGMRC 2118,BGMRC 2123,BGMRC 2134,BGMRC 2156,BGMRC 2138,BGMRC 2145,BGMRC 2177,BGMRC 2220 had significant inhibitory activity against Sporisorium scitamineum.The results showed that the sponge Pseudoceratina sp.associated bacteria in Xieyang Island had a rich diversity and high antibacterial activity,which provided a theoretical basis for exploring microbial resources in Xieyang Island and developing antibacterial active substances.
