Amorphophallus bulbifer refers to a group of konjac which have aerial bulbils in leaves.It has strong adaptability to high temperature and humidity environment as well as the strong resistance of soft rot disease and southern blight diseases.Partial species of A.bulbifer can produce apomictic seeds,which have a better retaining of the heredity of female parent.In recent years,it has become a widely planting species in konjac.At present,the study of A.bulbifer is restricted.In this study,apomictic characters were identified in A.bulbifer,which can provide theoretical support for the konjac industry.Amorphophallus muelleri was found to produce asexual seeds formation.Additionally,the seeds of A.muelleri were treated using gibberellin,the percentages of seed germination and the changement of the inclusions were analyzed during seed germinating.At last,starch synthesis related gene AmAGP was cloned from A.muelleri and its preliminary function was discussed.The main results were listed as follows:1.A.mwelleri produces apomictic seeds.In this study,eight species of bulbil were used to identify the character of apomixis.The results showed that A.muelleri displayed a high percentage of apomixis,which has a huge utilization in production.2.Gibberellin promote seed germination in A.muelleri.Effect of gibberellin on seed germination were discussed in this paper,the most suitable concentration of gibberellin for seed germination was observed in A.muelleri.In addition,the changes of the inclusions in seeds during germination were also analyzed.The results revealed that gibberellin promoted the decomposition of organic material in the seed and improved seed germination in A.muelleri.3.Clone of housekeeping gene 18S in A.muelleriIn this study,housekeeping gene 18S was cloned from A.mueller,the full-length cDNA of it was 841bp.It was homology with calla lily(AF168829.1).Studying for the housekeeping gene can provide theoretical support for further study.4.Clone and sequence analysis of ADP-GLUCOSE PYROPHOSPHORYLASE(AmAGP)in A.muelleri.Starch synthesis related gene AmAGP was cloned in A.muelleri,The length of the cDNA was 1660 bp,it encoded 552 amino acids.It was homology with Amorphophallus albus(JF727266.1)and Amorphophallus konjac(AF316326.1).AmAGP protein contained two conservative domains,transferase(NTP)and LbH-G1P-AT-C,and it located in the cytoplasm.In the secondary structure of protein,the percentage of random curl structure was more higher,it was 57.05%,followed by alpha helix,up to 21.56%.5.Expression levels of AmAGP in different tissues of A.muelleri.The temporal and spatial expressions of AmAGP were checked with semi-quantitative and Real-time PCR using corn,root,petiole,base of petiole and leaf.The results showed that AmAGP constitutively expressed in all tissues tested,its transcript level was high in corn and root.
