Zebrafish(Danio rerio)as an animal model,is widely used in genetics,neurobiology,developmental biology,drug screening,environmental monitoring work and other scientific research.However,the study that pathogens affect the zebrafish health status is relatively few.Especially bacterial disease which can caused high incidence,high mortality,severity of loss,is the focus of the research.In the current experimental,fish bacterial disease research is still relatively scarce,we launched study of this experiment,not only carry out a quality assurance for the laboratory,but also accumulate of scientific information for the model animal zebrafish to prevent and treat bacterial disease,which can achieve standardization of experimental fish testing and application provide a certain reference in the future.In this study,we isolated the pathogens named as No.2,No.3 and No.4 bacteria from infected zebrafish with typical pathogen and severe pathogenicity in the laboratory.We identified the bacteria with morphological observation and staining,physiological and biochemical characteristics,16S rDNA sequence analysis and other identification,and the result is No.2 bacteria belongs to Acinetobacter beijerinckii,No.3 bacteria belongs to Aeromonas veronii and No.4 bacteria belongs to Aeromonas media.In order to understand the pathogenicity of these three pathogens,the normal zebrafish were infected with different concentrations of pathogens.We observed and recorded the state of zebrafish within one week of treatment.The semi-lethal concentrations of A.beijerinckii,A.veronii and A.media were detected as 1.0×108cfu/ml?3.3884 × 1O5cfu/ml and 3.4674 × 106cfu/ml respectively,which the toxicity of A.veronii to zebrafish is the strongest.In order to eliminate the interference of pathogens to zebrafish,and to ensure the health of the experimental fish,we selected a common sterilization drugs for the three kinds of pathogens to do drug sensitivity test.The results showed that these three pathogens were extremely sensitive to lomefloxacin,levofloxacin and norfloxacin.Therefore,in the laboratory intensive breeding of zebrafish,these drugs can be used to control and prevention the diseases caused by pathogens.Through the study of this experiment,we have a basic understanding of the bacterial infection of zebrafish in the laboratory,which would provide the reference to ensure the quality of experimental fish.
