Objective:To investigate the expression of NF-kB and UBC 9 proteins during the process of differentiation from neural stem cells into neuron-like cells in Mouse and then study the expression of NF-kB and UBC 9 proteins in vivo during development of the nervous system in mouse embryos.Methods:Fetal brain-derived neural stem cells(NSCs)were isolated at embryonic day(E)11.5,and were induced..The expression of NF-kB and UBC 9 proteins during differentiation was detected by immunocytochemistry.In addition,Spinal neural tube-derived NSCs isolated at E12.5 were induced and detected in the same condition.Immunohistochemistry would be adopted to detect the expression of NF-kB and UBC 9 proteins at E9.5?E13.5?E15.5 and E18.5 during the development of embryonic spinal cord.Results:The expression of two proteins was tested positive during differentiation process of brain-derived NSCs,however,it showed descent with the extension of differentiation.The two proteins were found positively expression during the differentiation of Spinal neural tube-derived NSCs at E12.5 and the expression was gradually increased during differentiation which was positive in state of stem cell,and weak at initial differentiation.Besides,it was determined that nuclear translocation did not occur to NF-?B protein.Expression of NF-kB and UBC 9 proteins during the development of embryonic spinal cord was gradually increased and the nuclear translocation did occur to NF-?B protein.Conclusions:It was different between the expression of UBC 9 and NF-?B proteins during the differentiation of brain-derived NSCs and neural tube-derived NSCs and further exploration would be needed to inquiry the specific mechanisms of it.Expression of two proteins during differentiation of Spinal neural tube-derived NSCs was consistent with the trend in vivo,however,the difference was that during in vivo development of NF-?B,nuclear translocation occurred.
