With the development of economy and the progress of technology,the global energy is shortage.The most important is fossil energy is running out in the future.The development of human society will be limited.To find a new kind of energy is a method to solve this problem.Mutation breeding has a lot of advantages,such as high efficiency,low cost,simple and practical.It has already been used widely.This thesis have studied the biologic effects irradiated the microalgae(No.9 Chlorella,No.31 Chlorella and Chlorella.sp)with He-Ne laser,Nd:YAG laser and semiconductor laser of different time gradient.After the microalgae were irradiated,they were screened for the bio-oil production.Then there are 18 strains of mutants were screened out.The mutants which were screened were culture many generations.By means of researching the bio-oil production,it is easy to find that No.9 Chlorella which is exposed to Nd:YAG laser 8min,No.31 Chlorella which exposured to He-Ne laser 4min and 12 min,Chlorella.sp 9 which is exposed to Nd:YAG laser 4min,have higher bio-oil production than control.At the same time,the result that measured the activity of NR and ACCase showed the activities of the mutants were risen except the No.31 Chlorella which is exposed to He-Ne laser 4min.So we can preliminary conclude,He-Ne laser was not affect the activities of NR and ACCase in No.31 Chlorella which is exposured to He-Ne laser 4min laser directly,but the last three mutants were directly affected.We chose 7 key genes which play significant roles in pathways of lipid biosynthesis to clone.They are BC,FATA,ACP,SAD,MCTK,FAD,KAS.The expression of these genes in the four mutants we screened is risen.It is many times higher than control.Contrast the expression of the genes with the bio-oil production,it can be easily find that the genes have direct effect on the form of bio-oil.There are four mutants were screened in this research,they are No.9 Chlorella which is exposed to Nd:YAG laser 8min,No.31 Chlorella which exposured to He-Ne laser 4min and 12 min,Chlorella.sp 9 which is exposed to Nd:YAG laser 4min.It is hoped the paper can have some help to the researchers in this field.
