Objective:In recent years,the incidence of chronic kidney disease is increasing rapidly,which seriously endangers human health.Renal fibrosis is the pathological reaction of kidney in various pathogenic factors such as inflammation and injury.It is a common pathological change of chronic kidney disease.It is an important factor to determine the development and prognosis of kidney disease.Therefore,the treatment of renal fibrosis is the key to the treatment of nephropathy related diseases.Dongzhimen Hospital Professor Wang Yaoxian,according to the long-term clinical experience has the following 4 prescription.Fang 1 is a soft and loose knot group including 3 flavors of seaweed,oyster and turtle shell.Fang 2 is a recipe for activating blood and removing stasis,including 3 flavors of Salvia miltiorrhiza,safflower and Ligusticum chuanxiong,and 3 prescription of soft backbone and activating blood and removing stasis,and 4 commonly used prescription groups,consisting of seaweed,oyster,turtle,astragalus,angelica,and 6 drugs.The previous study of animal pharmacological experiments in Dongzhimen hospital found that the anti renal fibrosis group could significantly reduce the urine protein content of renal fibrosis rats,regulate its blood creatinine and urea nitrogen level,and have the effect on improving renal function of renal fibrosis rats.The method of serum pharmacology to further study its medicinal ingredients in the four prescription.Methods:1.Chemical and physical studies on promoting blood circulation to dissipate blood stasis and softening hard and dissipating stagnationTaking the 3 aqueous extract of Fang 3 as the research object,the liquid phase of 0.1%acetic acid water and acetonitrile was used as the mobile phase,and 300nm was selected as the detection wavelength and gradient elution,and the liquid phase Atlas of the group 3 was obtained.On the basis of the 3 characteristic map analysis,the HPLC-MS/MS analysis technology was applied to qualitatively analyze the chemical constituents in the compound.2.A study of common chemical composition in clinicTaking group 4 water decoction as the research object,with 0.1%acetic acid water solution methanol as the mobile phase,254nm was selected as the detection wavelength and gradient elution,and the group 4 compound liquid phase atlas was obtained.On the basis of the analysis of 4 liquid chromatogram,the HPLC-MS/MS analysis technology was applied to qualitatively analyze the chemical constituents in the compound.3.Study on serum drug chemistry of group 3Taking the rat serum as the research object,the HPLC-MS/MS liquid chromatography-use analysis technique was used to establish the chemical liquid chromatogram of the rat serum,and compared with the previous water extract to find the effective material basis.4.Study on serum drug chemistry of group 4Taking the rat serum as the research object,the HPLC-MS/MS liquid chromatography-use analysis technique was used to establish the chemical liquid chromatogram of the rat serum,and compared with the previous water decoction to find the effective material basis.Results:1.A lquid chromatogram of anti kidney fibrosis series group 3(Huoxue + Rujian)was established.2.A liquid chromatogram of anti kidney fibrosis series group 4 has been established.3.21 chemical constituents of group 3 of anti renal fibrosis group were qualitatively analyzed,of which 6 compounds were derived from Salvia miltiorrhiza,12 from safflower and 5 from Ligusticum chuanxiong,of which there were 3 common components in Ligusticum chuanxiong and safflower.4 chemical components that may play a role in serum were identified,which laid a solid foundation for clinical research.4.A qualitative analysis of 16 chemical constituents in the series of anti fibrosis kidney group 4 was made.Among them,13 compounds were from Astragalus,3 chemical components and angelica.We found out 4 possible chemical components in serum,which lay the foundation for later research.Conclusion:After 3 gavage,compounds were found in the serum of rats.Compounds Yi,Y2,Y3,may be the 3 chemical constituents,of which Y1 is vanillin(Vanilla acide);Y2 is salvianolic acid B(Salvianolic acid);After 4 gavage,compounds were found in the serum of rats.Compounds M1,M2,M3,and M4 may be the 4 Chemical Constituents of each side,of which Mi is Ligusticum lactone(Senkyunolide),M2 is Butylphthalide,M3 is Succinoadenosine,M4 is Formononetin. |
