Abstract/Summary:
Stellera chamaejasme is a dry rhizome of Stellera chamaejasme Linn.,belonging to the Stellera genus of the Thymelaeaceae family.mainly distributed in northern and southwest China.It is a traditional poisonous Chinese herbal medicine,originally contained in Shennong’s Herbal Classic of Materia Medica,whose nature and flavour is bitter and plain,can expel phlegm and kill insect.Up till now,photochemical studies on Stellera chamaejasme have shown that the presence of flavonoids,lignans,coumarins,diterpenoids,volatile oils and some other constituents.Modern pharmacological studies revealed that Stellera chamaejasme possessed a variety of bioactivities,such as antitumor,antibacterial,antiviral,anticonvulsant,antiepileptic and other aspects.Although Stellera chamaejasme has many pharmacological activities,its research focuses on the level of crude extracts,and monomer compounds are rarely reported.Therefore,it is very important to further study the chemical compounds of Stellera chamaejasme in order to better illuminate the material basis of it.In this dissertation,the chemical constituents of ethanol extract of Stellera chamaejasme were systematically isolated and structurally identified,and some of the isolated compounds were subjected to preliminary anti-cancer,anti-inflammatory,anti-oxidation,neuroprotection and hypoglycemic activities,which provides a scientific basis for the further development and utilization of Stellera chamaejasme.Thirty-three chemical compounds have been isolated by Silica gel,ODS,Sephadex LH-20 column chromatography and preparative HPLC.The structure of them were determined by extensive spectroscopic analysis including UV,IR,MS,HRESIMS,1D NMR,2D NMR,CD,ECD.These compounds included 9 lignans,2 sesquiterpenoids,6 diterpenoids,8 flavonoids,4 coumarins,and 4 other types of compounds.They are named as follows:stellerachamin A(1*),8-hydroxypluviatolide(2),wikstromol(3),pinoresinol(4),matairesinol(5),dextrobursehernin(6),hinokinin(7),(-)-glaberide I(8),medioresinol(9),sterallanol A(10*),auranticanol A(11),jolkinolide B(12),17-hydroxyjolkinolide B(13),gnidimacrin(14),stelleralide A(15),stelleralide F(16),stelleralide H(17),apigenin(18),chamaechromone(19),chamaejasmine(20),7-methoxychamaejasmine(21),neochamaejasmin A(22),isoneochamaejasmin A(23),Ruixianglangdusu B(24),chamaejasmine B(25),umberllliefrone(26),scopoletin(27),daphnetin(28),daphnoretin(29),palmarumycin SC1(30*),p-hydroxy ethylbenzoate(31),2,4,6-trihydroxy ethylbenzoate(32),syringic acid(33)。Among them,1,10 and 30 were new compounds,2,12 and 13 were obtained from the Thymelaeaceae family for the first time.The biological activity screening showed that Compound 29 possessed significant antitumor activity and Compounds 3,22,25,29,32,and 33 existed moderate neuroprotective effects.
