Objective:This study aims to analyze the relationship between acupuncture and aortic arch and glucose metabolism in cerebral functional regions in spontaneously hypertensive rats(SHR)using gene chip techniques.Methods:Experiment 1:effect of acupuncture at Ren Ying(ST 9)on gene expression profile of SHR aortic arch.20 spontaneously hypertensive rats were randomly divided into 2 groups,.They were Ren Ying(ST 9)group and model group.10 WKY rats were used as blank control group.The rats in the Renying(ST9)group received acupuncture at the bilateral acupuncture points for 20 minutes at noon every day,and rested one day every six days.The blank group and the model group did not carry out other interventions except for the same grasping and binding irritation.The systolic blood pressure of each group was measured at 1st,5th,9th,13 th,17 th,21 th,25 th and 28 th days after acupuncture intervention.After 4 weeks,the rat aortic arch 2-3mm was cut and RNA was extracted,and the Affymetrix whole genome expression chip was used to detect(P<0.05,fold change≥1.5 as the screening criteria).The function and pathway of gene enrichment related to the pathological process of hypertension were analyzed by GO enrichment and KEGG pathway enrichment,and the mechanism of the action of acupuncture on the blood pressure of Ren Ying(ST 9)was discussed from the angle of gene.Experiment 2:regulation of acupuncture on mRNA expression of PPAR-a and PPAR-βin SHR aortic arch tissue.The PPAR-α and PPAR-β gene were selected from the Ren Ying(ST 9)group VS model group and the model group VS blank group of the model group as the key genes.They were closely related to the incidence of essential hypertension and the regulation of blood pressure by acupuncture,and the mRNA expression of the two genes was verified by real-time quantitative fluorescence quantitative PCR.The changes in the expression of two genes mRNA in the aortic arch of SHR after the treatment of the model group and acupuncture were used to explore the molecular mechanism of the improvement of aortic vascular remodeling and SHR hypertension by acupuncture at the acupuncture point.Results:Experiment 1:in this experiment,gene chip results showed that in the aorta,the analysis of gene data in the model group showed 452 up-regulated and 245 down-regulation compared with the blank group.Compared with the model group,there were 261 up-regulated and 986 down-regulation in the model group.After acupuncture,we found that when we combined a large database of genes,we found that the frequency of genes was different and involved in biological regulation,metabolic process,cell biological process,catalytic activity,molecular binding,receptor activity,structural molecular activity,transport activity,cell component and macromolecule complex formation,etc.A biological process.The KEGG pathway analysis showed that the gene expression difference between the blank group and the model group was related to 12 pathways,and the genes that expressed differences between the human and model groups were related to the 10 pathways.Experiment 2:compared with the blank group,the expression of PPAR-a and PPAR-β in the model group decreased significantly(P<0.01).Compared with the model group,the expression of PPAR-a and PPAR-β in the human group was significantly increased(P<0.01,P<0.05),and the results of PCR were in accordance with the results of the chip.It is suggested that the mechanism of acupuncture regulating SHR blood pressure may be related to regulating the expression level of PPAR-a and PPAR-β in hypertension related genes.Conclusion:1.acupuncture can obviously reduce the blood pressure level of SHR,and its mechanism may be related to the differential expression of PPAR-a and PPAR-P in the aortic arch before and after acupuncture intervention.2.,acupuncture at Renying(ST9)can influence the gene expression profile of the SHR aortic arch.3.The abnormal expression of mRNA in the key gene of 3.SHR aortic arch may be an important molecular biological mechanism in the pathogenesis of essential hypertension.The effect of acupuncture on the blood pressure and target organ protection may be realized by regulating the expression of key genes and the signal transduction pathway.
