Background:Malt is a processed product of the Hortineur vulgare L.after germination and drying.In the clinic,raw,fried,and coke products are widely used.Raw malt could soothes the liver and enhanced the spleen,eliminates food and through milk;While fried malt could restrict milk and eliminates food;Coke malt,digestion and stagnation.Jiao malt,Jiao hawthornand Jiao Shen Qu are collectively known as Jiao Sanxian,which is commonly used in the treatment of diabetes.In recent years,researches on a-glucosidase inhibitors have been deepened,especially on natural Chinese herbal medicines.However,there are few studies on traditional carbon Chinese medicines,and there are few reports on the material basis of the Hypoglycemic effect of Jiaosanxian,and it is even unclear about the mechanism.To solve this problem,this study draw support the research methods of carbon dots in modern nanomaterials disciplines.And study its hypoglycemic efficacy and mechanism of action from a new perspective to explore the mystery of maltose.Purposes:Through combing the research on the mechanism of malt and diabetes,based on animal and in vitro experiments found that the basic substance of maltose reducing sugar and its effective mechanism.(1)By setting different temperatures of the muffle furnace,malting charcoal of different temperatures is prepared;(2)Taking advantage of modern nanomaterials research methods,carbon nano-components in malt charcoal with different temperatures are separated and purified;(3)The differences of a-glucosidase suppression activity of carbon malt in vitro were compared under different temperature conditions.(4)The malt charcoal with the optimum temperature is selected,and the malt carbon points in it are separated and characterized;(5)Establishing animal and in vitro experiments to study the material basis of the maltose reducing action of malt charcoal,and to make a preliminary exploration of the mechanism of carbonation of malt charcoal.(6)Provide new ideas and methods for obtaining new a-glucosidase inhibitors.Methods:(1)Preparation and conditions optimization of malt carbon dots:Malt charcoal with different temperatures was processed using a muffle furnace,and the malt components in the charcoal malt were separated using a nanomaterials method.And the malt charcoal processed under different temperature conditions was prepared.The components were characterized,and the pharmacodynamic study method was used to select the appropriate temperature to provide the foundation for the next study.(2)Composition comparative study of Malt and Charcoal Malt:In order to exclude the disturbance of other small molecules,the separated charcoal carbon dots were compared with those in malt.A simple comparison of the differences in the components of the malt and charcoal malt extracts after dialysis was carried out using HPLC method.(3)Characteristics of charcoal carbon dots:Using the research techniques and methods of nanomaterials to identify the charcoal carbon dots.Using ultraviolet-visible spectrophotometer(UV-vis),Fourier transform infrared spectroscopy(FT-IR),X-ray photoelectron spectroscopy(XPS),high-resolution transmission electron microscopy(HR-TEM)and other commonly used analytical methods in the field of research to study the hypoglycemic effect of malt carbon carbon.Based on short-term animal experiments,the effect of malt carbon spot on postprandial(oral sucrose or glucose)blood glucose was studied.(4)Preliminary study on the hypoglycemic effect of carbon in charcoal malt:A preliminary study of the mechanism of malt-charcoal carbon spot hypoglycemic effect was initially explored,and its hypoglycemic effect on a-glucosidase were preliminarily evaluated by in vitro a-glucosidase inhibition assay.Results:(1)Preparation of nano-components in charcoal malt and optimization of conditions:This experiment concocted charcoal malt at different temperatures,and was isolated and characterized.Under the electron microscope,the morphology of the malt charcoal extracts at different temperatures showed different dialysis,mainly in terms of particle size distribution.Infrared spectroscopic studies also found that the surface group species at different temperatures.The difference was obvious.The results of the study showed that malting charcoal processed at 300? had more carbon dots in morphology and group type.By studying the charcoal carbon points of malt processed under different temperature,it is preferable that malting charcoal carbon dots processed at 300?.with better hypoglycemic effect.(2)Comparative study of malt and malt carbon nano-components:In this experiment,the differences in the components of malt extract water and raw malt water extract were analyzed by high performance liquid chromatography.After the malt was charcoalized and dialysis to remove the small molecule components,the small molecule component was not shown in the HPLC results,but the components in the raw malt under the same conditions were complex.(3)Characterization of charcoal malt carbon dots:The analysis of malt charcoal's dialysate by means of UV-visible spectrophotometer,Fourier infrared spectroscopy,and projection electron microscopy which showed that the malt carbon dialysate showed uniformly distributed spheres under the electron microscope.The particle size distribution was between 2-8nm,lattice spacing 0.265.IR results show that the substance is rich in groups,there are many groups such as-OH,-COOH.UV-Vis spectrophotometer shows that it has weak UV absorption.(4)Study on the hypoglycemic effect of carbon in malt charcoal:After animal meal(orally sucrose or glucose)blood glucose experimental study,the results show that the malt carbon point in the intraperitoneal injection mode has a certain role in reducing postprandial blood glucose(5)Preliminary study on the mechanism of malt-induced blood glucose reduction in carbon malt:Inhibition of a-glucosidase activity in vitro,indicating that malt carbon dots significantly inhibit a-glucosidase activity,which has a hypoglycemic effect on charcoal malt.Conclusion:In this experiment,nanometer material research methods were used to separate and extract malt carbon dots from malt charcoal.Optimized the efficacy and characterize malt carbon dots through the in vitro a-glucosidase activity inhibitory experiment to,300? was selected.The conditions are suitable for the study.After the animal's postprandial(orally sucrose or glucose)blood glucose test,the malt carbon dots has the effect of reducing postprandial blood glucose,but after long-term administration,the mice have no effect on blood glucose and body temperature.The body weight of mice was significantly increased.Through the inhibition experiment of a-glucosidase activity in vitro,the mechanism of hypoglycemic action was initially studied in relation to the inhibition of a-glucosidase activity,and it was confirmed that the carbon point of malt and a-glucosidase activity became certain.This study provided an explanation for the hypoglycemic effect of charcoal malt on its material basis and its mechanism of action.At the same time,it also provided a new method and idea for exploring a-glucosidase inhibitors.This experiment uses the research methods of nanomaterials to provide ideas and methods for exploring the material basis of pharmacodynamics of Chinese herbal medicines.
