The diversity of natural scenic spots provides suitable environment for ticks.Ticks as the transmission medium of a variety of pathogens that transmission by sucking blood.Many of them are important zoonosis,which are harmful to the health of humans and animals.The purpose of this study is to investigate ticks and pathogens carried by ticks in five natural scenic spots in Harbin,so as to provide evidence for prevention and control of ticks and tick borne diseases.Part one: Investigation on distribution of ticks in five natural scenic spots in Harbin.6098 free ticks were collected application flags from five natural scenic area in Harbin.Dermacentor spp.(3955),Haemaphysalis spp.(1193)and Ixodes spp.(950)by morphological identification.A total of 2993 female ticks were collected,accounting for 49.08% of the total ticks and 50.92%(3105)of male ticks.According to the sampling points,species and gender,16 S rDNA sequence of 1056 ticks were amplified.These ticks were found to be 3 genera and 4species.They are Dermacentor silvarum(576),Ixodes persulcatus(240),Haemaphysalis japonica(239)and Haemaphysalis conicinna(1).Part two: The investigation of tick borne pathogens in five natural scenic spots in Harbin.Including high-throughput sequencing to investigat bacteria and applicat routine PCR to detect some pathogens.First,we applied high-throughput sequencing to sequenced the V3-V4 hypervariable region of 16 S ribosomal DNA for 1056 bacterial ticks.There are many species of ticks carrying bacteria in the 5 scenic spots,including a number of zoonotic bacteria,such as Brucella and Mycobacterium.Many tick borne bacteria,such as Coxiella,Rickettsia,Anaplasma,Borrelia and Ehrlichiawere detected too.It is found that there are some differences in bacteria at different locations.There are also significant differences among the different species.The abundance of the pathogens in the female is higher than that of the male.Secondly,we used routine molecular biology method to detect some important ticks borne pathogens in 1056 ticks.Borrelia and Babesia were detected.The detection rate of Borrelia spp was 8.05%(85/1056)and divided into 2 genotypes,which were Borrelia garinii 61.18%(52/85)and Borrelia afzelii 38.82%(33/85).Babesia caballi is only Babesia that detection rate of was9.38%(99/1056),while the detection rate of Theileria sinensis was 2.56%(27/1056).In summary,tick is existence in the five natural scenic spots in Harbin.Dermacentor silvarum are dominant ticks.The result of pathogens detection revealed that ticks carried some related diseases in these areas.The first hand data of ticks and tick borne diseases in somenatural scenic spots in Harbin can be mastered through this survey,which can provide reference for tick and tick borne diseases in these areas.
