| Purpose:To explore the effect of Huayu decoction on the inflammatory factors of diabetic macrovascular disease,and to further clarify the mechanism of Huayu Quzhuo traditional Chinese medicine compound on diabetic macrovascular protection.Material and method:Take SPF grade,Wistar rats,male 80,ten rats were randomly selected as normal control group according to body weight,given to the general feed;Randomly selected 10 as AS group,given high fat diet;And then randomly selected 12 as a traditional Chinese medicine prevention group,given high-sugar high-fat diet;The remaining 43 were fed through high-sugar,high-fat feed.After 30 days of feeding,the pathological model of diabetic rats with macroangiopathy was reproduced by intraperitoneal injection of streptozotocin and then divided into 6 groups according to the stratification of blood glucose level.The rats were divided into normal group,model group,Positive medicine group,traditional Chinese medicine prevention group,traditional Chinese medicine treatment group and AS group,each group of 10,a total of 60 rats were included in the experimental observation.Administration and feeding methods: Chinese medicine prevention group in the administration of traditional Chinese medicine at the same time should also be given high-sugar high-fat diet;positive drug group to give fosinopril sodium tablets for gavage;Chinese medicine treatment group given Chinese medicine gavage;AS group,normal group and model group were given equal dose of normal saline.The other groups were given the general diet,but the Chinese medicine prevention group was given high glucose and high fat diet.The rats were fed once a day for 8 weeks.During the experiment,rats were free to drink water.The levels of serum TNF-a and IL-6 in the rats were measured before and after treatment.The effects of traditional Chinese medicine on blood glucose and blood lipid were observed.The arterial endothelium,Vascular smooth muscle hyperplasia.Results:1.Huayu Quzhuo Chinese medicine can make DM rats blood glucose before modeling prevention group rats blood glucose had no significant difference with other groups;model of success and treatment of rat DM(except group AS)blood glucose level compared with the normal control group,except AS group,the rats of blood glucose the differences were statistically significant(P < 0.01),the model group blood glucose value was the highest;after traditional Chinese medicine treatment,compared with model group rats blood glucose were significantly decreased(P < 0.01),but it is not down to the normal range,after that compared the blood glucose decreased most obviously is Chinese medicine prevention group;positive drugs without obvious hypoglycemic effect(P > 0.05)was not statistically significant;the normal group compared with AS group,AS group rats blood glucose slightly increased,but the difference was not significant(P > 0.05)was not statistically significant.TG,TC and LDL-C decreased;traditional Chinese medicine prevention group and model group of diabetic macrovascular disease compared to traditional Chinese medicine prevention group can reduce TNF-a,IL-6 content and inhibition of TNF-a expression and IL-6 expression;light microscope showed that traditional Chinese medicine prevention group on arterial endothelial integrity and effective protection,but also has inhibitory effect on vascular smooth muscle hyperplasia.Conclusion:1.Remove stasis dispelling turbidity of traditional Chinese medicine compound can improve the experimental diabetic vascular disease in the rat's general status;2.Remove stasis dispelling turbidity of traditional Chinese medicine compound can improve diabetic vascular disease of the blood sugar,blood lipid disorders;3.Remove stasis dispelling turbidity of traditional Chinese medicine compound can inhibit the inflammatory factor TNF-a,the expression of IL-6;4.This study traditional Chinese medicine can protect diabetic vascular disease of the artery endothelial integrity,inhibition of vascular smooth muscle hyperplasia;5.This study traditional Chinese medicine of DM rats,blood vessels have a protective effect,can be big vascular disease prevention and control of DM rats. |
