Purpose: Through the establishment of severe acute pancreatitis(SAP)rat model of SAP rat intestinal epithelial cells promote the dynamic changes of anti inflammatory cytokine IL-6,IL-10 and MUC2 protein at different time points,given the intervention under the blood preparations to investigate the protective effect of Qing Qing,under the Huoxue Decoction on intestinal mucosa of SAP rats barrier injury and the related mechanism, to provide experimental basis for clinical treatment of severe acute pancreatitis.Material and method: (1)SD rats were randomly divided into sham operation group(blank group,n= 12),SAP group (model group,n=24),clearing and activating blood group(treatment group, n=24),each group was divided into 24h,48h,72h time points.(2)the blank group was only open, turning the pancreas, duodenum, intestinal and other organs after the closure of the abdomen. SAP group was treated with retrograde injection of 5% sodium taurocholate(0. 1mL/100g)into the pancreatic duct to prepare SAP rat model.After treatment,the treatment group was given 2H(1mL/100g),and then the rats were treated with 12h. (3) the rats in the three groups were collected after anesthesia at the time of 24, 48 and72h at the time of the operation. The blood was collected from the abdominal aorta, and the pancreas and the small intestine of the rats in the three groups were taken for detection. The serum amylase levels of rats were detected, the pathological changes observed in pancreatic and intestinal mucosa of rats, respectively, in accordance with the Schmidt standard and Chiu score method to evaluate the degree of pancreas and intestinal mucosa damage, quantitative fluorescence method (Rt-PCR) to detect the expression of intestinal epithelial cells in IL-6mRNA and IL-lOmRNA, the expression of MUC2 immunohistochemical method to detect intestinal epithelial cells in. SPSS 17.0 statistical software was used for analysis.Results:1.The pancreatic tissue was obviously swelling and hyperemia, which indicated that the model was successful.2.Qing Xia Huoxue Decoction can reduce SAP rat intestinal epithelial cells and promote the level of inflammatory cytokines IL-6, improve the level of inflammatory cytokines IL-10,reconstruction between proinflammatory and anti-inflammatory cytokines balance,reduce intestinal tissue injury.3.In the model group and the treatment group, the pancreatic pathological score at 24,48, and h was higher than that in the blank group (P < 0.01). The pathological score of pancreas in the treatment group at each time point was lower than that in the model group (P<0.05).4.The pathological model group and treatment group in 24, 48, three 72h time points of small intestine were higher than that of control group (P < 0.01); treatment group three time pathological score lower than the model group. The difference was statistically significant (P<0.05).5.The model group at each time point of the level of IL-6mRNA increased significantly compared with control group (P<0.05), and the increase degree is proportional to the time, but there was no statistically significant difference; compared with the model group, the treatment group at each time point IL-6mRNA level lower than the model group, the difference was statistically significant (P<0.05). Compared with the control group, the model group at each time point IL-10mRNA level increased significantly (P<0.01), and the increase degree and time is inversely proportional to (P<0.05); compared with the model group, the treatment group at each time point IL-10mRNA level increased significantly (P<0.05 or P<0.01),4872h, two time point is the most obvious.6. Compared with the blank group, the MUC2 staining score of the model group at each time point was significantly reduced (P<0.01); the treatment group and the model group, the MUC2 staining score at each time point increased, with statistical difference (P<0.05).Conclusion:1.Clearing away the blood circulation can reduce the serum amylase level and the score of pancreatic tissue in SAP rats.2.clear the blood circulation can reduce the SAP rat intestinal epithelial cell proinflammatory cytokine IL-6 levels, increase the level of anti-inflammatory cytokines IL-10, the promotion of proinflammatory and anti-inflammatory cytokines balance, reduce intestinal tissue damage.3.Clearing away the blood circulation can effectively protect the intestinal epithelial cells of SAP rats, increase the secretion of MUC2, so as to repair the intestinal mucosa, which can protect the intestinal mucosal barrier caused by SAP.
