Purpose:To explore the mechanism of Qing Xia Huo Xue Fa on hepatic injury of severe acute pancreatic in rats model by measuring Qing Xia Huo Xue Fang on severe acute pancreatitis rat serum interleukin-6,interleukin-10,aspartate aminotransferase,alanine aminotransferase,serum amylase and liver tissue expression level of nuclear factor kappa B expression.Material and method:The 60 SD rats,male and female,weight 180-220 g,were randomly divided into three groups:sham operation group(n=12),model group(n=24)and treatment group(n=24).The biliary pancreatic duct retrograde injection of 5% sodium taurocholate(0.1mL/100g)model in rats with severe acute pancreatitis.Sham operation group and acute pancreatitis rats fed with normal saline,and every 12 h into treatment group,rats were cleared under Huoxue granule(0.5mL/100g),and every 12 h was injected into normal diet.Each group were taken before12 h to stop drug and feed in for the first time after intragastric administration of 24 h,48h,72 h in 3 batches death.Morphological changes were observed in rat pancreas and liver tissue.The liver and pancreas tissue in 10% Formaldehyde Solution fixed,HE staining of liver and pathological changes of pancreatic tissue and score.Abdominal aortic blood serum was extracted and measured Serum ALT,AST,AMY were detected by ELISA.Serum IL-6 and IL-10 were detected by immunohistochemistry.The expression of NF-kappa B in liver tissue was detected by immunohistochemical staining.The above results were analyzed by SPSS19.0 software.Results:1 The pathological changes of pancreas and liver were normal in the sham operation group.The pathological damage of pancreas and liver in treatment group was lighter than that of model group at each time point.2 Model group and treatment group of AMY,AST,ALT concentration level in the same period were significantly higher than the sham group.Treatment group compared withmodel group at the same time,AMY,ALT and AST levels were significantly decreased(P <0.05).The serum IL-6 of the model group and the treatment group was significantly higher than that of the sham operation group at the same time,the treatment group was lower than the model group(P < 0.05).The serum IL-10 of the model group and the treatment group was significantly higher than that of the sham operation group at the same time,the treatment group was higher than the model group(P < 0.05).3 Sham operation group liver tissue in different time showed a small amount of NFkappa B expression,the expression of the model group and the treatment group of NF-?B in the same period of time compared to the sham operation group were significantly increased,the treatment group and model group compared to the same period,the expression of NF-?B were decreased(P < 0.05).Conclusion:1 Qing Xia Huo Xue Fang may adjust the anti-inflammatory cytokine balance in the treatment of severe acute pancreatitis.2 Qing Xia Huo Xue Fang can play a important role in protecting liver tissue by inhibiting the expression of NF-kappa B and reducing the damage of liver cells.
