Nowadays,the male infertility becomes commoner.There are many families facing the reprductive problems.Buslufan as one of the most widely-used chemotherapy drugs,can combinate with the DNA,biochemical enzymes and other biomacromolecule in the cells,and destroy the normal cell cycle to make the cell aoptosis or necrosis,so that the spermatogenic function was effected.To study the drugs for the male infertility,models treated by busulfan can be considered the classic model for drug study.Wuziyanzongfang is one of the famous Chinese medicines for treating male infertility.Accroding to clinical research,this medicine can obviously improve the sperm quality,and increase the male reproductive abilities.However,the mechanism is still not clear.Through the model of spermatogenic impairment treated with busulfan,this experiment tries to study about active ingredients of single herb of the Wuziyanzongfang on the spermatogenic function of the mice with spermatogenic impairment,and explore its mechanism preliminarily.1.The preliminary study about active ingredients of single herb of the Wuziyanzongfang on the spermatogenic function of the mice with spermatogenic impairment.In order to compare the effect of active ingredients of single herb of Wuziyanzongfang on the mice with spermatogenic impairment,this experiment 35mg/kg busulfan was injected intraperitoneally into 6.5-week Kunming male mice.After 40 days,the mice were given active ingredients of single herb of the Wuziyanzongwan with low-dose,middle-dose and high-dose respectively for 70 days.Observe the basic performance,body weight,weight of reproductive organs,sperm number and the histomorphology of testes.(1)basic performance:The mice in the spontaneous group shows the“shenxu”s symptoms,such as skin and mucous membrane pale,afraid of cold and reduced activities.Wuziyanzong-wan,mixed active ingredients of single herb,lycium barbarum polysaccharide and dodder polysaccharide can effectively treat the“shenxu”symptom in the mice with spermatogenic impairment.Raspberry ketone,schisandrin and psyllium saponins also can improve the"shenxu" symptom,but not effectively as others.(2)body weight:Compared to the normal group,the body weight of the spontaneous group is decreased obviously.The body weight in the group of 3 kinds of doses of mixed active ingredients of single herb,middle and high dose of Wuziyanzongwan,dodder polysaccharide and lycium barbarum polysaccharide,high dose of raspberry ketone and schisandrin are increased compared to spontaneous group.(3)the weight of reproductive organs:Compared to the normal group,the weight of the reproductive organs of the spontaneous group is decreased obviously.The testicular organ indexes in the group of,3 kinds of doses of mixed active ingredients of single herb,middle and high dose of Wuziyanzongwan,dodder polysaccharide and lycium barbarum polysaccharide,high dose of raspberry ketone are increased,similar to the results of epididymis organ indexes.(4)epididymal sperm motility and sperm number in epididymis:Compared to the normal group,the epididymal sperm motility and sperm number in epididymis of the spontaneous group is decreased obviously.3 kinds of doses of each group of active ingredients of single herb of the Wuziyanzongwan can improve the epididymal sperm motility;3 kinds of doses of lycium barbarum polysaccharide,dodder polysaccharide,Wuziyanzongwan and mixed active ingredients of single herb can improve the sperm number in epididymis.(5)sperm production in testicular tissue:Compared to the normal group,the sperm production in testicular tissue of the spontaneous group is decreased obviously.3 kinds of doses of lycium barbarum polysaccharide,dodder polysaccharide,Wuziyanzongwan and mixed active ingredients of single herb and middle and high dose of Wuziyanzongwan can improve the sperm number in testicular tissue.(6)The histomorphology of testicular tissue:Compared to the normal group,the The histomorphology of testicular tissue of the spontaneous group is different obviously,have little recover compared to the model.Wuziyanzongwan and mixed active ingredients of single herb>lycium barbarum polysaccharide>dodder polysaccharide>raspberry ketone>schisandrin>psyllium saponins.These results show that after two spermatogenesis cycles,spontaneous group only have little recover;among active ingredients of single herb of the Wuziyanzongwan,the effect of lycium barbarum polysaccharide is the best,and then dodder polysaccharide,raspberry ketone,schisandrin and psyllium saponins.2.The study of the effect of lycium barbarum polysaccharide on the spermatogenic function of the mice with spermatogenic impairment.Through the preliminary study about active ingredients of single herb of the Wuziyanzongwan on the spermatogenic function of the mice with spermatogenic impairment,we found that the group of lycium barbarum polysaccharide is better than other active ingredients of single herb,in terms of basic performance,body weight,the weight of reproductive organs,epididymal sperm motility and sperm number in epididymis,sperm production in testicular tissue and the histomorphology of testicular tissue.In order to further explore the menchanism of the effect of Wuyanzongfang,this chapter choose the group of lycium barbarum polysaccharide to study its effect on spermatogonial stem cells.Through immunohistochemical staining,study the expression of c-kit in mice testcle tissue.Through H-score and image processing by Image-Pro Plus,we got the number of positive cells and integrate optical density(IOD),and found that the expression of c-kit in the natural recovery group was lower than that in the 3 kinds of dose in LBP group and the normal control group.These results show that 3 kinds of doses of lycium barbarum polysaccharide can increase the level of c-kit in mice testicular tissue,and improve the development of spermatogonial stem cells.3.The study of the effect of lycium barbarum polysaccharide on the antioxidant ablity.The former two chapters found that the lycium barbarum polysaccharide can improve the spermatogenic function on mice with spermatogenic impairment,and increase the spermatogonial stem cells.This chapter through testing the SOD,GSH and other oxidant indexes to study the antioxidant ablity of lycium barbarum polysaccharide on mice testicular tissue,and explore the mechanism preliminarily..Extraction of frozen testicular tissue,homogenate,centrifuge and extract the supernatant as sample to be tested.Test the SOD through NBT colour method;test theThe GSSG,GSH and total glutathione through TNB colour method..We found that 3 kinds of doses of lycium barbarum polysaccharide can improve the level of antioxidant indexes in mice testicular tissue,such as SOD,glutathione.These results show that lycium barbarum polysaccharide contain the antioxidant ablity.Conclusion:?all active ingredients of single herb of the Wuziyanzongwan have an effect on the recover of mice with spermatogenic impairment,;the effect of lycium barbarum polysaccharide dodder polysaccharide and raspberry ketone are better than other active ingredients of single herb.?3 kinds of doses of lycium barbarum polysaccharide can increase the level of c-kit in mice testicular tissue,and improve the development of spermatogonial stem cells.?3 kinds of doses of lycium barbarum polysaccharide can improve the level of antioxidant indexes in mice testicular tissue,such as SOD,glutathione.These results show that lycium barbarum polysaccharide contain the antioxidant ablity.Background:The measurement of the age of the first spermatorrhea can be used to evaluate the growth and development of adolescent males to a certain extent.Through the analysis of the first spermatorrhea age,we can further discover the factors that affect adolescents adolescence.At present,China has a large number of reports on the first spermatorrhea age,but few comprehensive literature analysis.This paper summarizes 74 published academic literatures published from 1980 to 2013,and analyzes the basic state,change and characteristics of the juvenile's first spermatorrhea age.Objective:To analyze the variation and characteristics of the age of the first spermatorrhea among Chinese adolescents.Methods:74 literatures published between 1980 and 2013,containing 192 data,were analyzed.The data included about 435 thousand people.Results:In the 34 years,the age of the first spermatorrhea in Chinese adolescents showed an abvious declining tendency(y=-0.0424x+99.129,R2 = 0.3268,P<0.01).The average age of the first spermatorrhea was 14.40,from 1980' s 15.09 to 13.78 in 2013,with a decline of 1.22 during the 34 years.Multi-level sensitivity test with variable sample size,indicates that the age of the first spermatorrhea in Chinese adolescents has tended to occur in advance evidently.The Analytic Hierarchy Process about the decades shows that the average age of the first spermatorrhea is 14.63±0.62 between 1980 and 1994 with a decline of 1.35,14.44±0.63 between 1995 and 2004 with a decline of 0.54,14.07±0.49 between 2005 and 2013 with a decline of 0.12,and during these periods it was negatively correlated with the ages.Conclusion:The first age of spermatorrhea shows a obvious decline during the 34 years(P<0.05),but the changing rate between 1995 and 2004 is smaller than it between 1980 and 1994(both P<0.05).what' s more,it has little change between 2005 and 2013.Overall,the changing rate between 1995 and 2013 is smaller.Conclusion:During the 34 years,from 1980 to 2013,the first age of spermatorrhea declined significantly.It was affected by different decades,areas,rural-urban difference and their interaction.
