TXYF,a famous traditional Chinese herbal medicine(TCHM)formula,is one representative prescription in treating diarrhea-predominant irritable bowel syndrome(D-IBS),a disease with high incidence worldwide.This prescription contains Rhizome Atractylodis Macrocephalae,Radix Paeoniae Alba,Pericarpium Citri Reticulatae and Radix Saposhnikoviae.It was carefully designed and the drugs were combined based up on accurate efficacy.The present study was performed with purpose of developing a simple,sensitive and also specific HPLC-MS/MS method to simultaneously quantify the major bioactive components in rat plasma and tissues.The detailed results of the investigation were presented as follows:1.The ultrasonic-assisted extraction conditions were optimized using response surface methodology to obtain maximum extractive contents of 7 chemical constituents.2.In this work,a reliable and powerful method using HPLC-DAD-ESI-MS/MS was established for chemical investigation on the extract of TXYF.Twenty three components including monoterpene glucosides,flavonoids,chromones,lactones,organic acids and steroidals in the formula were successfully identified based on retention time,DAD and MS spectra compared with those of standards or literature data.The established method was successfully applied to determine the 7 components in real samples.3.A Triggered dynamic MRM(TdMRM)method is introduced here,in which MRM is performed in two ways to simultaneously quantification and confirmation.The dynamic MRM allows the instrument to acquire primary MRM transitions only during a stated retention time window,thus reducing the number of concurrent ion transitions.Importantly,this dramatically shortened MS scan time is kept constant In addition,the secondary transitions are triggered automatically when the intensity of the primary transitions exceed a preset threshold(Triggered MRM).4.11 constituents of TXYF were simultaneously determined in rat plasma/brain tissue/urine using a novel TdMRM algorithm.The TdMRM method provides higher sensitivity than conventional MRM.
