Depression is a syndrome of emotional disorder as a mental sickness,which characterized by a prolonged decline in mood.T It has a higher incidence of morbidity,disability,and suicide,and has the characteristics of depression,anxiety,mental damage,loss of appetite and insomnia.Thus become a harmful disease to human health.According to WHO publicly available statistics show that depression is ranked as the world's fourth highest disease.The average incidence of global depression is estimated to be about 11%,which is expected to be in 2020,that is,within the next five years,will become second only to heart disease;the top second diseases.In China,the incidence of depression is about 3%?5%,there are more than more than 2600 people suffer from depression.Recent epidemiological studies have shown that the lifetime prevalence of depression can reach 16%,ranking the highest in China's disease burden of disease by up to second.There is a lack of specific methods to provide the perfect solution for depression in diagnostics.Treatment give first place to western medicine therapy and electroconvulsive therapy.The treatment of acupuncture therapy on the treatment of depression has good performance in safety,effectiveness and antidepressants compared with western medicine.Acupuncture has a rapid onset,relatively minimal side effects and cost as a series of advantages.There has been a large number of research results for the mechanism of acupuncture treatment of depression,but it has not been able to fully explain the onset of the target and all the pathway.Further research is needed.ObjectiveThis study attempts to clarify the effect of electro acupuncture on the treatment of depression through the behavior observation of electroacupuncture on chronic stress rat model of depression,and using molecular biology methods to further explore the mechanism of the expression of PDE4D PDE4A potential target.Methods1.Animal grouping The animals were randomly divided into four groups(n=8):blank group,control group,drug group,electroacupuncture treatment(EA)group.Rats in each group were fed for one week before the experiment.Then the chronic stress stimulation and different intervention were carried out.The rats in the blank group were fed with the group,the other groups were isolated.Each group of rats were fed in different isolation room.2.Modeling method The rat model of chronic stress depression was established by chronic stress combined with isolation support by reference.Control group,drug group and EA group received 21 days of 7 different stimulations,including swimming in cold water,no water,night shift,clipping tail,bound,humid environment.Stimulations were randomly assigned to each day at an average rate of 3 times.3.Treatment methods:Blank group:fed in the normal conditions for 21 days,no isolation,no stress stimulation;Model group:chronic stress for 21 days,with the same bound as EA group;Drug group:chronic stress with isolation,1 mg/1ml in distilled water to prozac.According to weight,the prozac was made before the 1h intragastric administration,once a day for 21 days;EA group:chronic stress with isolation,EA treatment for 21 days,and stress stimulation was 1h after the daily EA.4.Detection methods of targets:Open Field Test,body weight and sucrose preference test was performed to test the behavior of rats.The expression of hippocampus PDE4A.PDE4D was detected by using Real-Time PCR and Western Blot.Results1.Behavioral changeThere was no significant difference in the behavior of the rats before the experiment(p>0.05).At the end of the experiment,Open Field Test,body weight and sucrose preference results of blank group were higher than those in model group(P<0.01).Drug group and electroacupuncture group rats were higher than that in the above aspects.Compare with model group,there was significant difference(P<0.01);In terms of sucrose consumption,compared with drug group,EA group has no significant difference(p>0.05);The sucrose preference percentage,compared with drug group,the percentage of sucrose preference in electroacupuncture group was higher than that of drug group,there was significant differences(p<0.05).2.Expression of PDE4A and PDE4D mRNA in hippocampusCompared with control group,the PDE4A,PDE4D mRNA expression of hippocampus of rats in the model group increased significantly(P<0.01),while in the electroacupuncture group rats the expression compared with the model group decreased significantly(P<0.01),and no significant differences between blank group(P>0.05).Compared with the blank group,there was no significant difference in the expression of hippocampus in drug group(P>0.05).3.Expression levels of PDE4A and PDE4D protein in hippocampusCompared with control group,the PDE4A,PDE4D protein expression of hippocampus of rats in the model group increased significantly(P<0.01).The expression of the drug group also increased,there was significant difference(P<0.05);There were significant differences in between electroacupuncture group and drug group(P<0.01).Conclusion1.Fluoxetine and electroacupuncture can improve the symptoms of depression.EAhas a more obvious advantage in improving the behavior of the lack of pleasure,and can improve the sensitivity of reward.2.PDE4A and PDE4D can be used as potential indicators of depression,which are associated with depression.3.Electro acupuncture can inhibit the expression of PDE4A and PDE4D mRNA in the PDE4 subtype.The effect of similar to PDE4 inhibitors on depression can be produced without the side effects of PDE4 inhibitors.4.Fluoxetine could not inhibit the expression of PDE4D protein,and EA had obvious advantages in inhibiting the expression of PDE4A and PDE4D protein.EA may be associated with depression by several single subtype PDE4 inhibition,and depression related to regulation of target more specific.This provides a better effect in the treatment of depression,which is one of the possible mechanisms to improve depressive state.
