| Psoriasis vulgaris,known as ’Baibi’ in Traditional Chinese Medicine,is a common recurrent chronic inflammatory skin disease genetic correlation mediated by T lymphocytes,with scaly erythema as the main clinical manifestations.Its etiology has not been fully elucidated.This disorder is difficult to completely cure,what-s worse,severe cases can affect skeletal and visceral system.The prevalence rate have obvious difference in the world population.As shown in studies,the Nordic is the highest,the epidemiological survey showed that the prevalence rate of psoriasis in China is rising.Long-term medication not only seriously affects the life quality of the patients,but also caused great social and economic burden.TCM treatment of psoriasis has certain advantages,the theory of Chinese medicine as the main blood disease,the use of heat-clearing and blood-cooling of traditional Chinese medicine,not only can relieve clinical symptoms of psoriasis recurrence,you can also adjust the patient’s constitution,so as to relieve psoriasis disease process.Based on TCM theory,’blood heat’ is the main cause of psoriasis vulgaris.According to this core pathogenesis,Professor Yan-ping Bai worked out the Heating-clearing and blood-cooling recipein,which was for patients with blood-heat syndrome psoriasis vulgaris.This recipein conforms to the pathogenesis and main characteristics of psoriasis pathogenesis characteristics,with clearing heat and cooling blood detoxification effect.Its clinical therapeutic effect has been confirmed by our rgoup via some clinical trials and animal experiments,especially for the upset,thirst and other symptoms are obviously improved.Through animal experiments it has showed obvious effects on inhibiting hyperplasia of epidermal cells,as well as promoting the formation of granular layer,thus inhibit psoriatic epidermal hyperplasia and improve parakeratosis.As the literature,activated T cells as a key effect on the pathogenesis of psoriasis,which can secrete a variety of cytokines,including IFN-y,IL-6 and so on.These cytokines will be transfered into the nucleus by JAKs/STATs,Notch,NF-κB,Wnt,MAPK signaling pathways,notably the JAKs/STATs pathway for the research focus in recent years.Suppressor of cytokine signaling(SOCS1)can inhibit the JAKs/STATs pathway,which has a positive effect on the treatment of psoriasis.Studies have shown that blocking the role of cytokines in keratinocytes can inhibit the excessive proliferation of keratinocytes and reduce the secretion of inflammatory cytokines,thus blocking the occurrence of psoriasis.To study the effect on biological characteristics of Heating-clearing and blood-cooling recipein via mouse model induced by imiquimod,revealing the upregulation of SOCS1 negative regulation of the JAKs/STATs pathway,reduce the secretion of cytokines which play a role in the treatment of psoriasis,will help to further explore the mechanism of psoriasis and rich the theory of TCM in treating psoriasis.Methods:The Imiquimod Cream was employed in this study to establish the animal model of psoriasis.The mice were divided into blank group,model group,Tripterygium wilfordii group,Heating-clearing and blood-cooling recipein group(low,moderate and high dose)and were given corresponding intervention for 8 days.During the course,we observe the morphological changes periodically.Histopathological HE staining and immunohistochemistry were performed to observe histological changes and the expressions of SOCS1,STAT3 and JAK1 in skin tissue of mice.Moreover,real-time quantitative PCR and Western Blot method were carried out to detecte the mRNA and proteins expression of SOCS1,STAT3 in signal pathway and IFN-γ,TNF-α,IL-6,IL-1.Results:(1)Using imiquimod cream on the back skin of BALB/c male mice,1-2mm thickness,twice a day,for 8 consecutive days,the mice skin showed symptoms like psoriasis,such as infiltrative erythema,thick layer of silver white scales,and some scattered change in the bleeding point.Their histological features showed hyperkeratosis with parakeratosis,acanthosis,dermis and perivascular infiltration of inflammatory cells,consistent with the pathological features of psoriasis.(2)The expression of SOCS1 in model mice spinosum cells increased compared with the normal group,Tripterygium wilfordii group and Heating-clearing and blood-cooling recipein with different concentrations of extract group reduced in different degrees compared with the model group,the low concentration group was least,Tripterygium wilfordii group,moderate and high concentration group increase respectively.STAT3,JAK1 in model mice spinosum cell nuclei in the deposition obviously,Tripterygium wilfordii group and different concentrations of Heating-clearing and blood-cooling recipein extract group than in the model group increased in different degree,the low concentration group was least,Tripterygium wilfordii group,moderate and high concentration group increased in turn.(3)The expression of SOCS1,STAT3,TNF-α,IL-6,IL-1,IFN-ymRNA in model group skin was increased compared with the normal group,the difference was statistically significant(P<0.05);the mRNA level of Tripterygium wilfordji group and different concentrations of Heating-clearing and blood-cooling recipein groups expression were statistically increased compared with the normal group(P<0.05);Compared with the model group was statistically decreased(P<0.05);among them,the high concentration group decreased most significantly,followed by Tripterygium group,moderate and low concentration group.(4)The expression of SOCS1,STAT3,TNF-a,IL-6,IL-1,IFN-yprotein in different concentrations of Heating-clearing and blood-cooling recipein were significantly increased compared with the normal group,the difference was statistically significant(P<0.05),the model group increased most significantly,followed by low,high concentration group in turn,the differences were statistically significant(P<0.05).Conclusion:(1)To use imiquimod cream on BALB/c male mice back skin,thickness 1-2mm,twice a day,for 8 consecutive days,can get the animal model that basically in accordance with the clinical and pathological features of psoriasis.(2)The secretion of TNF-α,IFN-y,IL-1 IL-6 and other cytokines increased in the lesions of model mice induced by Imiquimod cream,which was in accordence with psoriasis to some extent.(3)Heating-clearing and blood-cooling recipein may reduce the secretion of TNF-α,IFN-γ,IL-1,IL-6 through activating the JAKs/STATs pathway,thus playing the role of healing psoriasis.Moreover,this effect in moderate and high dose group were more obvious than the relatively low group. |
