Objective:To establish rat models of allergic contact dermatitis(ACD)and to explore the mechanism of Qinlou Qingli Pill.Method:1 Forty SD rats were randomly divided into blank group(only dorsal hair removal in two places).model group(only establish the ACD model),treatment group(establish the ACD model and apply Qinlou Qingli Pill),and positive control group(establish the ACD model and apply Cetirizine Hydrochlorde Tablets).The ACD model was established by 0.1%of DNCB solution in the first and second hair removal department,and then treated with medicine after the second stimulation.2 Evaluating skin efficacy and observing the pathological changes of the skin by HE staining.The concentration of IL-4,TNF-a and LTB4 in serum were detected by enzyme linked immunosorbent assay(ELISA).The mRNA expression of BLT1,Caspase-14 in skin lesion were tested by Real Time-Polymerase Chain Reaction(RT-PCR).Results:1 Animal experiment results show that:The edema,erythema,papules,moss exfoliation degree reduced significantly among the rats of treatment group and positive control group,the total effective rate was 90%,and there is significant difference compared with the model group.2 HE staining show that:the skin of the back of the rats in the model group was mild hyperkeratosis,and a large number of inflanmatory cells infiltrated in the epidermis and dermis.While in positive control group and the treatment group,the stratum comeum became thinner,and the infiltration of inflammatory cells in epidermis and dermis is significantly lower than that in the model group.3 Comparison of LTB4 and BLT1:The results showed that the serum levels of LTB4 in the skin lesion of model group is significantly higher than that of the blank group(P<0.01),while in the treatment group and positive control group the levels of LTB4 decreased significantly than that of model group,treatment group(P<0.01),positive control group(P<0.05);The PCR results showed that the expression of BLT1 mRNA in the skin lesion of model group were significantly lower than that of the blank group(P<0.01);while in the treatment group and positive control group BLT1 mRNA were significantly increased,however,there had no statistical difference between the two groups(P>0.05).4 Comparison of TNF-a and IL-4:The results showed that the serum levels of TNF-aof model group were significantly higher than that of the blank group(P<0.01),while the levels of TNF-a in the treatment group and positive control group decreased significantly,positive control group(P<0.01),treatment group(P<0.05),and the results of the two groups showed no apparently difference(P>0.05);The serum levels of IL-4 in each groups had no significant difference(P>0.05).5 The level of Caspase-14:The results showed that contents of Caspase-14 mRNA in the skin lesion of model group were significantly higher than that of the blank group(P<0.05).while in the treatment group and positive control group the level decreased,treatment group(P<0.05),positive control group(P<0.05);and the results of the two groups showed no apparently difference(P>0.05).Conclusion:1 Qinlou Qingli Pill could inhibit the LTB4-BLT pathway and reduce the local inflammatory response.2 Qinlou Qingli Pill can reduce the serum level of TNF-a of ACD rats.3 Qinlou Qingli Pill could be beneficial to the skin barrier function,which had achieved the effects of treating allergic dermatitis.
