Effect Of SangLongZhiYang Pill On Treatment Of Pruritus And Modulation Of Levels Of Itching Mediators

Objective:To observe the treatment effect of SangLong ZhiYang pill on pruritus and to explore the anti-pruritic mechanism.Methods: Including two parts. The first, 131 cases with chronic eczema or neurodermatitis, who were up the selected sta ndard of cases and that pruritus was their principle action, were divided randomly into the test group treated with SangLongZhiYang pill and the control group treated with WuSheZhiYang pill. The survey was conducted on clinical effect after a period of therapy, scores of dermatology quality of life and content of itching mediators such as 5-HT, SP, B-END in serum or plasma before and after treated. The second, effects of SangLong ZhiYang pill on itching threshold value with histamine in guinea pig and levels of itching mediators such as interleukin-2(IL-2)in serum of DTH mice, leukotriene B4 (LTB4)produced and released in rat polymorphonuclear leukocytes (PMNs) were studied.Results: 1.Clinical observation showed that, SangLongZhiYang pill had agood clinical curative effect on chronic eczema and neurodermatitis that the total effective rates were respectively 87.10% and 92.31%, but, in the control group treated with Wu SheZhiYang pill, the total effective rates were only respectively 62.96% and 73.53%, comparison between the two groups showed significant statistical difference (P<0 05or P<0.01).2 . Both of SangLongZhiYang pill and WuSheZhiYang pill could significantly improve dermatology quality of life scale of patients with chronic eczema or neurodermatitis, and the effect of improvement with SangLongZhiYang pill was remarkably better than that with WaSheZhiYang pill(P<0.05).3.Clinical experiment studies showed that, both of SangLongZhiYang pill and WusheZhiYang pill could significantly reduce content of itching mediators such as 5-HT, SP and B-END in serum or plasma of patients with chronic eczema and neurodermatitis. Eut also, the reduced level of the test group was remarkably better than that of the control group(P<0.05or P<0.01).4.The animal experiment results of different dose (high, middle, lower)test groups of SangLongZhiYang pill all shoved that SangLongZhiYang pillcould raise the itching threshold value with histamine in guinea pig, reduce the content of IL-2 in serum of DTH mice and inhibit the LTB4 product in rats PMNs. Among which, the function of modulation was better when middle or high dose was given. And more, there was certain relation between dose and function which SangLongZhYang pill inhibited the LTB4 product in rats PMNs(R=0.99).Conclusion: SangLongZhiYang pill had a remarkable chinical effect on pruritic dermatoses such as chronic eczema and neurodermatitis. In these patients with pruritic dermatoses, there was an abnormal increasing of the level of itching mediators such as 5-HT, SP and P -END.The anti-pruritic mechanism of SangLongZhiYang pill may be correlated with suppression or resistance of itching mediators such as 5-HT, SP and B-END.
