| Diabetes has become a major threat to human health and is considered one of the most common chronic diseases.It is a systemic disease based on glucose metabolism disorders.According to the latest data of the International Diabetes Federation,there are 463 million people with diabetes in the world aged 20-79,of which type Ⅱ diabetes accounts for 90-95 %.It can be seen that type 2 diabetes seriously affects human’s health.The drugs including insulin secretion,biguanides,α-glucosidase inhibitors and thiazolidinediones can regulate T2DM significantly in clinical practice.Nevertheless,most of drugs which were adopted to treat the T2DM catholically have some side effects including weight gain,gastrointestinal irritation,inducing diarrhea,nausea and vomiting.Therefore,it is important to research a novel natural hypoglycemic product that are safe and without toxic side effects.Gynura procumbens(GP),an edible herb,have good hypoglycemic effect in T2DM.However,the molecular mechanism of GP on T2DM is still unclear.Based on this,the paper carries out the following research: 1.Study on network pharmacology of Gynura procumbens in type 2 diabetes mellitusAs the appearance of the concept of multi-target drug discovery and in-depth studies of network biology,network pharmacology emerged.It is a powerful means which can be used to explore the function and behavior of a sophisticated biological system systematically.It has altered the definition of drug discovery from "one target,one drug" to "multi-targets,multi-components therapy." It has irreplaceable advantages in the research of TCM and has been widely used in the molecular mechanism research of TCM.The molecular mechanism of GP to treat T2DM was predicted based on the network pharmacology technology.Finally,we obtained 37 potential targets and 10 signaling pathways.This study laid the foundation for the the molecular mechanism research of GP in T2DM.2.Study on the hypoglycemic activity of Gynura procumbens in IR-HepG2 cellsThe model of IR-HepG2 cells was established to study the hypoglycemic activity of GP.The results indicated that GP(1.25 mg/mL)had prominent hypoglycemic effect.Additionally,the result of RT-PCR and western-blot suggested that GP regulate the crucial genes and proteins significantly in the PI3K/Akt signaling pathway.These results showed that GP may have significant hypoglycemic activity in vitro and provided a foundation for further studies of vivo.3.Study on type 2 diabetes mellitus treatment by Gynura procumbens in C57BL/KsJ-db/db miceDue to the complex and diverse etiology and ethical reasons for human diabetes,experimental research on the pathogenesis,complications,and drug evaluation of diabetes must rely on animal models to provide an important basis for human etiology and therapy of diabetes.This experiment discussesd the therapeutic effect of GP on T2DM based on db/db mice.Firstly,behavioral indicators including blood indicators,liver,pancreas,kidney and other pathological indicators of mice were tested.Moreover,RT-PCR and western-blot methods were adopted to explore the mechanism of GP in T2DM and results showed that GP can significantly alleviate the crucial genes in PI3K/Akt signaling pathway.Similarly,these consequences suggested that GP regulated T2DM diabetes in vivo and provided a theoretical basis for the further development and research of GP.In this work,the method of network pharmacology was applied to speculate the potential targets and potential signaling pathways of GP in hypoglycemic activity.IR-HepG2 cells and T2DM mice were selected as experimental models to investigate the molecular mechanisms of GP against hyperglycemia.These studies provide new insights and theoretical basis for the further research and application of GP. |
