[Objective]1.Through high sugar,high fat feed,feed rats,combined with intraperitoneal injection:chain urea rhzomorph streptozotocin(STZ),The blood glucose,blood lipid,liver-related transaminase,liver morphology,structure and function were tested to confirm whether the model of diabetic rats with liver injury could be established successfully.To observe the effects of electroacupuncture at "Zhenyu",electroacupuncture "non-acupoint" and glimepiride on blood glucose,blood lipid,body weight,liver-related transaminase,liver morphology,structure and function of diabetic rats with liver injury.And compared the three intervention methods,the initial judgment,electroacupuncture "pancreatic Yu" points on the liver of diabetic rats have a protective effect.2.the expression of glucagon-like peptide-1(GLP-1)protein in the liver of diabetic rats was detected,And the difference of gene expression and protein expression between the blank group and the model group,the difference of the expression level after intervention,To detect the expression of GLP-1 gene and protein in the liver,and to explore whether the electroacupuncture at "Pancreas" can protect the liver injury in diabetic rats.[Methods]Forty male rats were randomly divided into two groups:control group(n = 8)and control group(n = 8).The remaining 32 rats were used as model group and fed with high glucose and high fat diet for 50 days.After one hour of intraperitoneal injection of STZ solution for 72 hours,random blood glucose,molded standard:random blood glucose>16.7mmol/L.Animals were divided into blank group,model group,electroacupuncture,"Pancreas"group,electroacupuncture"non-acupoint" group,"glimepiride drug" group,all animals 8/cage feeding.Among them,the blank group in the adaptive feeding 1 week after the end of the machine extraction,the rest of the animals in the adaptive feeding after modeling and evaluation of the mold,the random extraction.The intervention was 1/d,6d/week for 4 weeks.Electroacupuncture parameters:continuous wave,5Hz,3-5mA,20min.All animals were sacrificed at the tail of the rats every other week,and the fasting blood glucose and body weight were measured in rats.The levels of serum alanine transaminase(ALT),aspartate transaminase(AST)and y-glutamyltransferase(GGT)were measured after the last intervention.The expression of GLP-1 in the liver was detected by real-time PCR(RT-PCR).The GLP-1 protein was detected by Western blotting(Western blot).[Results]Experiment 1:take high sugar,high fat forage diet,combined with intraperitoneal injection of STZ solution,create type 2 diabetes(type 2 diabetes mellitus;T2DM)rat model,ChengMo standard:random blood glucose or greater tendency for 16.7/L,OGTT experiment 2 h blood glucose acuity 11.1 mol/L.Statistics show that blood sugar,each time point model group were higher than blank group(p<0.05),the difference was statistically significant;Model group rats total cholesterol,total cholesterol,TC),low-density lipoprotein cholesterol,low density lipoprotein,cholesterol,LDL-C);,three acyl glycerin(triglyce:ride;TG)levels were significantly higher than that of blank group(p<0.05),HDL-C was significantly lower than that of blank group(p<0.05).the difference was statistically significant.Model group rats of AST,ALT,the GGT levels were significantly higher than that of blank group(p<0.05),the difference was statistically significant.And HE staining hepatic biopsy:liver cells showed extensive swelling,sinus gap significantly narrowed,inflammatory cell infiltration and fibrous tissue hyperplasia,visible,there is an obvious damage to the liver of rats.Curative pancreatic shenshu point,can quickly adjust the rise in blood sugar in type 2 diabetes rats,its long-term effect is more reliable,and is better than that of electric acupuncture an acupuncture point group.At the same time,the curative pancreatic shenshu point,also can restrain rapid weight loss in type 2 diabetic rats,but with the progress of the course of the disease,its effect in regulating weight is not significant.However,for each index of blood fats,the curative pancreatic shenshu point,can effectively reduce the type 2 diabetic rats by elevated levels of TC,TG,LDL-C,and on the adjusting TG,LDL-C,beauty is better than that of Glenn urea to fill the stomach;And on adjusting the TC,LDL-C,including superior to the hole.In addition,the curative pancreatic shenshu point,can effectively reduce the increase in type 2 diabetic rats serum levels of AST,ALT,GGT.and its effect is better than that of electric acupuncture an acupuncture point.Curative pancreatic shenshu point,can effectively reduce the increase in type 2 diabetic rats serum levels of AST,ALT,GGT,and its effect is better than that of electric acupuncture an acupuncture point.Liver pathological section:the curative pancreatic shenshu point,can effectively reduce liver cell cytoplasm red dye,nucleus pycnosis,cell apoptosis,inflammation cells infiltration,small bile duct hyperplasia,vesicles lipid drops of the phenomenon,improve lipid metabolism disorders,and to improve the effect,is better than that of electric acupuncture of acupuncture point group and Glenn urea,cupping the den and Glenn urea group,also can be a very good improved inflammatory cells infiltration and small bile duct hyperplasia,and Glenn beauty urea improving lipid vesicles dripping phenomenon,including superior to the acupuncture point group.Experiment 2 shows:1.in the liver of rats showed the presence of glp-1 receptor protein expression,and appear in 53 kD protein bands,same as the other experimental proof.Grey value of liver glp-1 receptor protein detection,after statistics analysis,comparable between groups(p>0.05),is standard object with blank group,model group,the pancreatic yu group,Glenn urea,the acupuncture point group of glp-1 receptor proteins grey value,a downward state(p<0.05);Compared with model group,pancreatic shu,Glenn urea group,the acupuncture point group still has significant increase(p<0.05);Compared with pancreatic yu group,Glenn beauty urea with the acupuncture point group glp-1 receptor proteins grey value,there was no statistically significant difference,while all lower than pancreatic yu group,but there was no significant difference;Compared with urea Glenn beauty group,the acupuncture point group of glp-1 receptor proteins grey value,higher than Glenn beauty urea groups,and pancreatic yu group are basically identical.2.1n the expression of gene level,the gene expression of model group,pancreatic group,non-acupoint group and glimepiride group was significantly lower than that of blank group(p<0.05),and the difference was statistically significant(p<0.05);Compared with the model group,the expression of gene expression in the Pancreas group was significantly higher than that in the model group(p<0.05),but there was no significant difference between the glibenclamide group and the non-(P<0.05).The combination of glimepiride group and non-acupoint group was similar to that of non-acupoint group(P<0.05).[Conclusions]It was suggested that the model of diabetic rats with liver injury was successfully established by intraperitoneal injection of high glucose and high fat diet and combined with STZ solution.The follow-up experiment was completed in accordance with the related literature.Electroacupuncture at the Pancreas,can effectively regulate blood glucose in type 2 diabetic rats,inhibit the weight loss of diabetic patients in the short term,significantly reduce TC,TG,LDL-C levels,and effectively improve the lipid metabolism disorders,and further protection.Electroacupuncture,"Pancreas" points,can effectively increase the GLP-1 receptor protein and gene expression.Protecting the liver may be through lowering blood glucose,blood lipids,improving glucose and lipid metabolism disorders,resulting in the role of increased GLP-1 receptor protein and gene expression,and ultimately improve diabetic liver injury.
