The first part: Effect of DingChuanTang on immune cytokines of respiratory syncytial virus bronchiolitis in ratsObjective:In this study,through the establishment of respiratory syncytial virus bronchiolitis model in young rats,Using Elisa method to detect the changes of immune cytokines(IFN-??IL-4?IL-10?IL-13?IL-33?IL-17)in serum of RSV infected rats,thus reflecting the mechanism of Th1/Th2 related cytokines imbalance after RSV infection,to further study the effect of DingchuanTang on cytokines in RSV infected rats,clarifying the mechanism of Dingchuantang in preventing and treating RSV infection,to reduce the incidence of asthma after RSV infection providing a scientific basis.Methods:1.36 Wistar rats were randomly divided into control group,RSV infection group,ribavirin group,DingchuanTang group,each group of 9,in addition to the control group,the remaining group infected with RSV(Long strain)virus intranasally,after intranasal,the rats in the control group and the RSV infection group were treated with equal volume of saline,each intervention group was given the corresponding drug by gavage.The effects of Dingchuantang on the prevention and treatment of RSV bronchiolitis in rats were evaluated by observing the general status,lung index and pathological changes of the rats in each group.2.Using Elisa method to detect the changes of immune cytokines(IFN-??IL-4?IL-10?IL-13?IL-33?IL-17)in serum of each group.Results:1.On the 7th day after infection,compared with the control group,the general status of rats and the pathological changes of lung tissue were significantly different between the two groups(P<0.05);compared with RSV infection group,the general condition and pathological changes of lung tissue in DingChuanTang group and ribavirin group were improved,and the lung index was significantly different(P<0.05);there was no significant difference between the ribavirin group and the DingchuanTang group(P>0.05).2.The results of the expression of cytokines in serum were shown:compared with the control group,the levels of IFN-y and IFN-y/IL-4 were significantly lower and the other indexes were significantly higher in RSV infection group than those in control group(P<0.05);compared with RSV infection group,the levels of IFN-?/IL-4 and IL-4,IL-10 and IL-13 in ribavirin group were significantly lower than those in RSV infection group(P<0.05),the levels of IFN-?and IFN-?/IL-4 in IL-4,IL-10,IL-10,IL-13,IL-33 and IL-17 were significantly lower than those in DingChuanTang group(P<0.05);Compared with the ribavirin group,the levels of IFN-y were significantly higher and IL-33 were significantly lower in DingChuanTang group than those in the ribavirin group(P<0.05).Conclusion:1.DingChuanTang can reduce the damage of lung pathological of RSV infection bronchiolitis,effectively prevent RSV infection.2.In RSV infected rats,Thl related cytokines decreased and Th2 related cytokines increased,inducing Th1/Th2 equilibrium drift.3.DingChuanTang can significantly inhibit the Th2 related cytokines reaction and enhance the Thl related cytokines reaction after RSV infection in rats,which has a two-way regulation effect on the imbalance of Th1/Th2 related cytokines after RSV infection.The second part:Study on the level of immune cytokines in children with bronchiolitisObjective:Study about changes of serum levels of immune cytokines(IFN-??IL-4?IFN-?/IL-4?IL-10?IL-13?IL-33?IL-17)in children with bronchiolitis from Th1/Th2 balance,to clarify the pathogenesis of bronchiolitis,to provide a theoretical basis for preventing bronchiolitis from developing asthma in the future.Methods:1.Using prospective research methods,from September 2016 to February 2017 collecting less than 2 years old children with bronchiolitis in XX Children's Hospital,Department of Respiratory Medicine as bronchiolitis group.At the same time,the healthy children of the same age were selected as the control group,and the changes of immune cytokines in the two groups were observed?2.Using Elisa method to detect the changes of immune cytokines(IFN-??IL-4?IL-10?IL-13?IL-33?IL-17)in serum of control group and bronchiolitis group.Results:The results of the expression of cytokines in serum were shown:compared with the control group,IL-4,IL-10,IL-13,IL-33 and IL-17 in the bronchiolitis group were significantly higher than those in the control group(P<0.05).Conclusion:The morbidity of bronchiolitis in children is closely related to changes of the level of cytokines,and the body is in Th1/Th2 related cytokines immune imbalance.
