There are over one million hectares of existing tea plantations in our country.The leaves,flowers and seeds are discarded as waste,except for the young leaves.It is the research on the tea leaves,branches and seeds for extraction and analysis of active material.At the same time,the anti aging experiment was carried out.It is expected that the research results can provide basis for the application of tea tree extracts in the fields of medicine,food and daily chemical products,promote the improvement of tea varieties,increase the value of tea production and the income of tea farmers and promote the greening of barren hills.1.Extraction and analysis of effective parts of tea plantSolvent extraction was used to extract the effective parts of leaves and branches and seed oil was extracted by aqueous extraction.Target components were identified by TLC.The content of index components was determinated by HPLC and the tea tree extract was analyzed.The yield respective of tea leaves extract,branches extract and seed oil is as follows:23.35%,14.08%and 19.73%.The leaves extract samples with EGCG and EC,branches with EC,seed oil with squalene shows the same color spots in the same position in TLC.The content of EGCG and EC in leaves extract is 12.010%,1.026%,the content of EC in branches extract is 0.677%and the content of squalene in seed oil is 0.820%measured by HPLC.2.Optimization of extraction technology for tea leaves by central composite design response surface methodologyEGCG content in leaves was used as the evaluation index.On the basis of single factor investigation,the extraction process of leaves was optimized by central composite design response surface method.Optimum extraction technology was as follow:extracted with 13.08-fold the amount of 68.84%ethanol for 60.34 minutes and the content of EGCG in leaves is 31.59 mg·g-1.3.Evaluation of anti-aging effect of tea tree extract by transdermal administrationAfter DPPH free radical scavenging assay to determine the antioxidant activity of tea extracts,ageing models of mice were established by injection of D-galactose combined with ultraviolet radiation.Tea extract was given by transdermal delivery.HE staining and Masson staining were used to observe the skin tissue,the biochemical indexes and the water content was detected in the skin.The results showed that tea tree extract had strong ability of free radical scavenging.Compared with the model group,the skin of mice in the control group were smooth,the thickness of epidermis and corium were thick,the collagen fibers were thick,long and neat,the content of SOD,Hyp and water of skin were raised while the content of MDA were decreased.4.Study on Preparation and quality of tea tree creamFour kinds of O/W cream prescription were designed and compared and determine the best prescription.The quality of tea tree cream was studied from the aspects of appearance,the value of pH,microbial limit,water absorption,spreading and washability,skin irritation test and content determination.Through the in vitro skin permeation test,the distribution of index component and permeation rate of cream in the skin was investigated.The proportion of each component of tea cream:7%sixteen alcohol,3%stearic acid,4%glycerol monostearate,3%seed oil,3%leaves extract,3%branches extract,15%glycerin and 62%water.The tea tree cream was determined to be O/W type,the value of pH is 6.48.The microbial limit,water absorption,spreading and washability,skin irritation of tea cream were in line with the requirements.By HPLC method,the content of EGCG was 0.391%,the content of EC was 0.0505%and the content of squalene was 0.0247%in the cream,and the linear fitting equation of EGCG accumulated infiltration and time in the unit area of the cream was Qn=2.2375t+5.3267(R2=0.9916).5.Evaluation of anti-aging effect of tea tree extract by oral administrationAgeing models of mice were established and tea extract was given by oral administration.The learning and memory ability of the mice was compared by water maze test in each group.The main results are as follows:Compared with the model group,the escape latency of mice in treatment group was short,the number of crossing platform was increased,time in platform quadrant becomes longer,the content of SOD in serum was increased obviously and the content of MDA was decreased significantly.
